Bronchopulmonary dysplasia

Alexander R. Schmidt et al.

Summary: Bronchopulmonary dysplasia is a common adverse outcome of prematurity, characterized by arrested lung growth, alveolar simplification, and pulmonary vascular dysangiogenesis in newer cases. Patients may present with impaired pulmonary function, reactive airway disease, or exercise intolerance, while the most serious long-term complication is the development of pulmonary vascular disease and pulmonary hypertension. Treatment often includes diuretics, steroids, bronchodilators, or oxygen supplementation, with additional perioperative anesthetic risk for children with pulmonary hypertension.

Summary: Bronchopulmonary dysplasia (BPD), as one of the major sequelae following premature birth, remains a significant challenge in terms of prevention and treatment. Despite some new scientific insights, the available therapeutic options are still limited.

Design-Based Stereology of the Lung in the Hyperoxic Preterm Rabbit Model of Bronchopulmonary Dysplasia

Christian Muehlfeld et al.

Summary: This study aimed to investigate the pathophysiological characteristics of bronchopulmonary dysplasia (BPD) using a rabbit model in a hyperoxic environment. The results showed that rabbit pups exposed to high oxygen environment had impaired lung function, signs of pulmonary hypertension, decreased alveolar number and area. Additionally, they exhibited thickened alveolar septa, intra-alveolar accumulation of edema fluid and inflammatory cells, as well as thickened walls and smaller lumen in nonparenchymal blood vessels.

Capillary Changes Precede Disordered Alveolarization in a Mouse Model of Bronchopulmonary Dysplasia

Svenja Appuhn et al.

Summary: The study conducted on neonatal mice exposed to different oxygen concentrations revealed that disrupted vascular development may lead to impaired alveolarization, supporting the vascular hypothesis of BPD. Alveolar capillary network in hyperoxia-exposed mice showed rarefaction and altered geometry, with effects on alveolar epithelium and capillary endothelium appearing at different time points. These findings provide valuable insights into the pathogenesis of BPD.

Lung ultrasound features and relationships with respiratory mechanics of evolving BPD in preterm rabbits and human neonates

Barbara Loi et al.

Summary: Comparative study of lung ultrasound and mechanical characteristics in preterm rabbits and human neonates with evolving BPD revealed that the preterm rabbit model can fairly reproduce the features of evolving BPD seen in clinical practice, with similar lung ultrasound findings and altered lung mechanics parameters. The study also demonstrated consistent relationships between ultrasound-assessed lung aeration and airway resistances in both the animal model and human neonates.

Patho-mechanisms of the origins of bronchopulmonary dysplasia

Mitali Sahni et al.

Summary: Bronchopulmonary dysplasia (BPD) remains a common complication of premature infants, characterized by impaired lung alveolarization and dysregulated vascularization. Recent research has focused on pathways of oxidative stress-related lung injury, placental insufficiency, growth factor signaling, and microRNAs in understanding the maldevelopment of the premature lung.
