CD8+ T cells and fatty acids orchestrate tumor ferroptosis and immunity via ACSL4

Peng Liao et al.

Summary: T cell-derived interferon gamma (IFN γ) in combination with arachidonic acid (AA) induces immunogenic tumor ferroptosis, serving as a mechanism for CD8(+) T cell-mediated tumor killing. This process involves IFN γ stimulation of ACSL4 and alteration of tumor cell lipid pattern.

Copper induces cell death by targeting lipoylated TCA cycle proteins

Peter Tsvetkov et al.

Summary: Copper is an essential cofactor for all organisms, but excessive copper can lead to cell death. We found that copper-dependent cell death mechanism is different from known mechanisms, and it is related to mitochondrial respiration. Copper directly binds to lipoylated components of the tricarboxylic acid cycle, causing protein aggregation and loss of iron-sulfur cluster proteins, ultimately leading to cell death.

Selective Targeting of Cancer Cells by Copper Ionophores: An Overview

Valentina Oliveri

Summary: Conventional cancer therapies face challenges due to off-target effects, leading to the need for new therapeutic agents with increased selectivity, reduced side effects, and the ability to overcome cancer cell resistance targeting cancer stem cells. Some copper ionophores show promise in inducing cuproptosis in cancer cells with greater selectivity.

Identification of a novel cuproptosis-related gene signature and integrative analyses in patients with lower-grade gliomas

Jia-hao Bao et al.

Summary: Cuproptosis is a newly discovered form of programmed cell death in lower-grade gliomas (LGGs), and its prognostic value and correlation with the tumor microenvironment (TME) have not been studied. In this study, we investigated the genetic and transcriptional variation, prognostic value, and expression patterns of cuproptosis-related genes (CRGs) in LGG. We developed a risk model based on the CRG score to predict LGG patients' prognosis and found that a high CRG score was associated with worse survival outcomes. The CRG score was also correlated with TME characteristics and treatment responses, suggesting its potential as a therapeutic target.

Prognosis and personalized treatment prediction in TP53-mutant hepatocellular carcinoma: an in silico strategy towards precision oncology

Chen Yang et al.

Summary: This study retrospectively analyzed 1135 HCC patients and developed a random forest-based prediction model to estimate TP53 mutational status, as well as a robust poor prognosis-associated signature which showed superior ability to predict survival in TP53-mutant patients. In silico screening revealed three targets and two agents that might have potential therapeutic implications in high-PPS patients, presenting a comprehensive view of potential treatment strategy.

The Immunomodulatory Potential of Copper and Silver Based Self-Assembled Metal Organic Biohybrids Nanomaterials in Cancer Theranostics

Neela Prajapati et al.

Summary: Copper high aspect ratio structures (CuHARS) and silver cystine nanoparticles (AgCysNPs) exhibit extensive anti-cancer effects on glioma tumor cells. CuHARS increases nitric oxide production to reduce glioma cell viability, while AgCysNPs demonstrate significant toxicity.

Methylation and Expression of the Exercise-Related TLR1 Gene Is Associated With Low Grade Glioma Prognosis and Outcome

Lichun Lu et al.

Summary: The study identified potential exercise-related genes, with TLR1 identified as a promising biomarker for predicting outcomes in low-grade glioma patients. Reduced expression and hypermethylation of TLR1 were associated with better survival outcomes, suggesting its potential as a prognostic marker for this cancer. Furthermore, TLR1 was found to be involved in immune cell infiltration and immunotherapy in LGG.

Relation of Serum Copper Status to Survival in COVID-19

Julian Hackler et al.

Summary: The study suggests that serum levels of copper and selenium in COVID-19 patients are related to survival rates, with higher concentrations of copper and ceruloplasmin associated with higher chances of survival. Additionally, copper and selenium levels in serum showed a positive correlation in contrast to expectations, indicating their importance in predicting survival and influencing disease course.

Novel Immune Infiltrating Cell Signature Based on Cell Pair Algorithm Is a Prognostic Marker in Cancer

Hao Zhang et al.

Summary: In this study, the infiltration pattern of 65 immune cells was collected and evaluated to construct the Immune Cell Pair (ICP) score, which showed a robust reliability and efficacy in predicting the survival of patients with gliomas, in pan-cancer samples, and six independent cancer types. The ICP score was also correlated with the genomic alteration features in gliomas.

Immune Infiltrating Cells-Derived Risk Signature Based on Large-scale Analysis Defines Immune Landscape and Predicts Immunotherapy Responses in Glioma Tumor Microenvironment

Nan Zhang et al.

Summary: The study systematically evaluated the TME infiltration pattern of 2877 glioma samples and established a TME scoring system. High TME scores were associated with increased immune infiltrating cells, elevated expression of immune checkpoints, increased mutation rates of oncogenes, and decreased survival rates of patients. The TME score may serve as a valuable prognostic biomarker for gliomas.

Siglec15 shapes a non-inflamed tumor microenvironment and predicts the molecular subtype in bladder cancer

Jiao Hu et al.

Summary: Siglec15 is specifically overexpressed in the tumor microenvironment (TME) of various cancers, likely designing a non-inflamed TME in bladder cancer (BLCA). High expression of Siglec15 in BLCA correlates with lower immune infiltration, lower response to cancer immunotherapy, and a higher incidence of hyperprogression. A combination of anti-Siglec15 and cancer immunotherapy may be a more effective strategy than monotherapy, while an immune risk score (IRS) could accurately predict prognosis and response to treatment.
