Small intestine and colon tissue-resident memory CD8+T cells exhibit molecular heterogeneity and differential dependence on Eomes

Yun Hsuan Lin et al.

Summary: Tissue-resident memory CD8+ T (TRM) cells are a subset of memory T cells that are important for controlling infection. TRM cells in the small intestine and colon exhibit heterogeneity in terms of cytokine and granzyme expression, transcriptional and epigenetic regulation, and functional characteristics. The transcription factor Eomes plays a context-specific role in supporting the maintenance of established TRM cells in the small intestine but not in the colon.

Tissue-resident memory CD8(+) T cells possess unique transcriptional, epigenetic and functional adaptations to different tissue environments

John T. Crowl et al.

Summary: This study investigates the diversity of gene expression and genome accessibility in mouse CD8(+) T-RM cells in different tissues, and identifies molecules critical for the generation of CD8(+) T-RM cells in response to acute viral infection. The research reveals both common and tissue-specific transcriptional and epigenetic signatures in T-RM cells from different organs. T-RM cells in the intestine and salivary glands express TGF-beta-induced genes and are maintained by ongoing TGF-beta signaling, while those in the fat, kidney, and liver do not. Furthermore, the study identifies the transcriptional repressor Hic1 as a critical regulator of T-RM cell differentiation in the small intestine, and shows that Hic1 overexpression enhances T-RM cell differentiation and protection from infection.

Pyroptosis-Mediated Periodontal Disease

Mariane Beatriz Sordi et al.

Summary: Pyroptosis is a pathological mechanism in the cause of periodontitis, involving caspase activation, cleavage of gasdermin D, and secretion of interleukins to induce tissue destruction. Removing periopathogens' virulence factors that trigger pyroptosis may be a potential strategy to combat periodontal disease.

Summary: The oropharyngeal mucosa serves as a primary site for pathogen entry and viral replication. During acute oral mucosal viral infection, ILC1s play a crucial role in providing early protection through IFN-γ production, enhancing resistance against oral mucosal viruses.

Association of Viral Infections With Oral Cavity Lesions: Role of SARS-CoV-2 Infection

Giusy Rita Maria La Rosa et al.

Summary: Oral lesions associated with COVID-19 have diverse etiopathogenesis including ulcerations, aphthous-like lesions, and macules, predominantly found on the tongue, lips, and palate. These lesions in COVID-19 patients may be underestimated, potentially linked to the direct or indirect action of SARS-CoV-2, coinfections, immune impairment, and drug reactions. Further research is needed to establish the diagnostic and pathological significance of oral manifestations of COVID-19.

CD8+ T Lymphocytes: Crucial Players in Sjogren's Syndrome

Huimin Zhou et al.

Summary: Primary Sjogren's syndrome (pSS) is a chronic autoimmune disease characterized by damage to multiple organs and glands, with dry eyes, dry mouth, and enlarged salivary glands being common clinical manifestations. CD8(+) T lymphocytes have been found to play a significant role in acinar injury in the exocrine glands, contributing to the pathophysiology of the disease. Targeting CD8(+) T cells in treatment strategies can offer promising new approaches for managing pSS.

Expansible residence decentralizes immune homeostasis

Sathi Wijeyesinghe et al.

Summary: The immune system in adult mice is integrated within tissues, with a durable maintenance of resident memory T cells contributing to tissue immunity. Tissue pliancy facilitates the accumulation of tissue-resident memory without eroding pre-existing antiviral T cell immunity, resulting in the decentralized homeostasis of organismal immunity.

SARS-CoV-2 infection of the oral cavity and saliva

Ni Huang et al.

Summary: The study highlights the potential role of the oral cavity in COVID-19 pathogenesis, with evidence of SARS-CoV-2 infection and transmission via saliva. Viral shedding dynamics in saliva correlated with symptoms, and asymptomatic individuals were found to harbor sustained salivary antibodies against SARS-CoV-2 post-recovery.

Local and systemic mechanisms linking periodontal disease and inflammatory comorbidities

George Hajishengallis et al.

Summary: Periodontitis is causally linked to the development of other chronic inflammatory diseases outside the oral mucosa, and intervention studies have shown that local treatment of periodontitis can ameliorate surrogate markers of comorbid conditions. Experimental animal studies have further strengthened the potential causal link by establishing biologically plausible and clinically consistent mechanisms. Understanding the impact of disseminated periodontal pathogens and systemic inflammation associated with periodontitis on extra-oral pathologies may lead to new therapeutic options to reduce the risk of periodontitis-associated comorbidities.
