Two divergent haplotypes from a highly heterozygous lychee genome suggest independent domestication events for early and late-maturing cultivars

Guibing Hu et al.

Summary: By studying the genome of lychee, it was found that lychee had undergone two independent domestication events in Yunnan and Hainan, China. The maturity differences between early-maturing and late-maturing lychee cultivars may be regulated by gene deletions. This research is important for understanding the domestication history of lychee and improving lychee and related crops.

A 3D gene expression atlas of the floral meristem based on spatial reconstruction of single nucleus RNA sequencing data

Manuel Neumann et al.

Summary: Cellular heterogeneity in growth and differentiation leads to organ patterning. Single-cell transcriptomics allows the study of gene expression heterogeneity in developing organs at an unprecedented resolution, but it loses the original physical location information of cells. This article proposes a method that combines single-nuclei RNA-seq with microscopy-based 3D spatial reconstruction to reconstruct the genome-wide gene expression patterns of individual cells in a 3D flower meristem, and to investigate gene expression differences among meristematic domains giving rise to different tissue and organ types.

FLOWERING LOCUS T paralogs control the annual growth cycle in Populus trees

Domenique Andre et al.

Summary: In temperate and boreal regions, perennials adapt their annual growth cycle to the change of seasons. Populus trees have evolved FT2 for regulating vegetative growth and dormancy, and FT1 is essential for spring bud flush, showing how FT duplication and sub-functionalization allow Populus trees to regulate different developmental control points.

Systematic Methods for Isolating High Purity Nuclei from Ten Important Plants for Omics Interrogation

Ming-Chao Yang et al.

Summary: Recent advances in developmental biology have been made possible by using multi-omic studies at single cell resolution. However, progress in plants has been slowed, owing to the tremendous difficulty in protoplast isolation from most plant tissues and/or oversize protoplasts during flow cytometry purification. Surprisingly, rapid innovations in nucleus research have shed light on plant studies in single cell resolution, which necessitates high quality and efficient nucleus isolation. The protocol outlined here makes it feasible to perform plant omic studies at single cell resolution.

Ning Zhai et al.

Summary: Single-cell profiling of callus from hypocotyl explants reveals the heterogeneity and gene networks crucial for plant regeneration, highlighting the essential roles of WOX5 and WOX7. The tissue structure and molecular interactions within the callus are similar to root primordium, with WOX5 promoting endogenous auxin production and interacting with key regulators to enhance pluripotency and organ regeneration.
