Transcriptome Profiling of ADAR1 Targets in Triple-Negative Breast Cancer Cells Reveals Mechanisms for Regulating Growth and Invasion

Allison R. Baker et al.

Summary: ADAR1 plays a critical role in triple-negative breast cancer, and its loss leads to reduced cell growth, invasion, and mammosphere formation. ADAR1 regulates both coding and noncoding targets by altering gene expression level, A-to-I editing, and splicing. The study also reveals that several tumor suppressor miRNAs are upregulated upon ADAR1 loss, suppressing cell-cycle progression and invasion.

ADARs act as potent regulators of circular transcriptome in cancer

Haoqing Shen et al.

Summary: This study demonstrates that ADARs play a significant role in regulating the circular transcriptome. ADARs can stabilize or destabilize circRNA secondary structures through editing, and also enhance the binding of RNA-binding proteins. These ADARs-regulated circRNAs are widely expressed in various cancers and have functional relevance to tumorigenesis.

SMILE Downregulation during Melanogenesis Induces MITF Transcription in B16F10 Cells

Xuan T. Truong et al.

Summary: SMILE is a transcriptional corepressor that plays a critical role in the regulation of melanogenesis. It is regulated by α-MSH/cAMP/protein kinase A signaling and acts by suppressing the transcriptional activity of the cAMP response element-binding protein. SMILE's decreased expression in melanoma tissues suggests its potential role as a corepressor in skin pigmentation.

RNA editing enzyme ADAR1 controls miR-381-3p-mediated expression of multidrug resistance protein MRP4 via regulation of circRNA in human renal cells

Yuji Omata et al.

Summary: This study reveals that downregulation of ADAR1 increases the expression of MRP4 in human renal cells. ADAR1 represses the biogenesis of circRNA circHIPK3 through its RNA editing activity, which interferes with miR-381-3p and induces the expression of MRP4.

Circular RNA circLMO7 acts as a microRNA-30a-3p sponge to promote gastric cancer progression via the WNT2/β-catenin pathway

Jiacheng Cao et al.

Summary: The study found that circLMO7 can promote the proliferation, migration, and invasion of gastric cancer cells by acting as a sponge for miR-30a-3p to affect the WNT2/beta-Catenin pathway. Animal experiments also showed that circLMO7 could promote gastric cancer growth and metastasis in vivo. Furthermore, circLMO7 can also impact the glutamine metabolism of gastric cancer cells through the WNT2/beta-Catenin pathway to enhance its malignant biological function.

Circular RNA circDLC1 inhibits MMP1-mediated liver cancer via interaction with HuR

Hailing Liu et al.

Summary: Our study demonstrates that circDLC1 is downregulated in HCC tissues and is closely associated with a favorable prognosis. Overexpression of circDLC1 inhibits proliferation and motility of hepatoma cells, while silencing of circDLC1 has the opposite effect. Mechanistically, circDLC1 binds to RNA-binding protein HuR, reducing the interaction between HuR and MMP1 mRNA, leading to inhibition of MMP1 expression and ultimately suppressing HCC progression.

Evaluating the therapeutic potential of ADAR1 inhibition for triple-negative breast cancer

Che-Pei Kung et al.

Summary: Triple-negative breast cancer (TNBC) is the deadliest form of breast cancer with no targeted therapy available. The ADAR1 enzyme, highly expressed in TNBC, is essential for the survival of TNBC cell lines. Knockdown of ADAR1 in TNBC cells attenuates proliferation and tumorigenesis, leading to translational repression and elevated IFN stimulated gene expression. These findings establish ADAR1 as a novel therapeutic target for TNBC tumors.
