Gut immune cell trafficking: inter-organ communication and immune-mediated inflammation

Sebastian Zundler et al.

Summary: Immune cell trafficking in the intestine plays a crucial role in the pathogenesis of immune-mediated inflammatory diseases. This Review discusses the mechanisms of immune cell trafficking along the gut-liver, gut-joint, and gut-brain axes, and provides an overview of their implications for treatment options in IMIDs.

Residual homing of α4β7-expressing β1+PI16+ regulatory T cells with potent suppressive activity correlates with exposure-efficacy of vedolizumab

Emily Becker et al.

Summary: This study investigated the non-linear exposure-efficacy characteristics of vedolizumab in IBD patients. The research found that blocking T-Eff cell trafficking with vedolizumab while allowing residual homing of T-Reg cells could be a strategy to optimize treatment outcomes. These findings were supported by data from Gemini II and III trials.

Etrolizumab-s Does Not Induce Residual Trafficking of Regulatory T Cells

Anna Schweda et al.

Summary: The efficacy of etrolizumab in inducing residual trafficking of regulatory T cells is not notable, which may explain the disappointing results in recent clinical trials. This suggests that the Teff overweight in the inflamed gut might persist despite reduced overall T cell recruitment.

Regulation and Role of αE Integrin and Gut Homing Integrins in Migration and Retention of Intestinal Lymphocytes during Inflammatory Bowel Disease

Mary E. Keir et al.

Summary: Targeting interactions between alpha 4 beta 7 integrin and endothelial adhesion molecule MAdCAM-1 to inhibit lymphocyte migration to the gastrointestinal tract is effective in treating inflammatory bowel disease. Upregulation of ICAM-1, VCAM-1, and MAdCAM-1 at gene and protein levels was observed in both ileal and colonic tissue from active IBD patients compared to healthy subjects. TGF-beta 1 treatment induced alpha E expression on T cells, suggesting a potential mechanism for upregulating alpha E beta 7 in response to TGF-beta within the gut mucosa of IBD patients.

Human Intestinal Macrophages Are Involved in the Pathology of Both Ulcerative Colitis and Crohn Disease

Suranga Dharmasiri et al.

Summary: Intestinal macrophages in patients with UC and CD exhibit cellular reprogramming, inflammatory phenotypes, and differences in key pathways. Macrophages in CD patients also display M2 characteristics, potentially exacerbating inflammation associated with CD-specific features.

Targeting Immune Cell Trafficking - Insights From Research Models and Implications for Future IBD Therapy

Maximilian Wiendl et al.

Summary: Inflammatory bowel diseases such as Crohn's disease (CD) and ulcerative colitis (UC) are multifactorial diseases with an increasing prevalence and incidence worldwide. Targeting immune cell homing has become an important pillar of IBD therapy, with potential for further therapeutic interventions. Leukocyte trafficking and tissue-derived chemokines offer untapped resources for potential therapeutic interventions in IBD.

Summary: Selectins and integrins play crucial roles in recruiting leukocytes to inflamed tissues. Studies have found that E- and P-selectins have different effects on the activation of α(L)β(2), with varying responses observed at different temperatures.
