Engineered virus-like particles for efficient in vivo delivery of therapeutic proteins

Samagya Banskota et al.

Summary: We have developed and applied engineered DNA-free virus-like particles (eVLPs) for efficient delivery of gene editing tools, achieving high levels of base editing. By overcoming the limitations of cargo packaging, release, and localization, the fourth-generation eVLPs demonstrated efficient base editing in mouse and human cells. In addition, the off-target effects of eVLPs were significantly reduced compared to other delivery methods such as AAV or plasmid.

Dynamic Polymer Amphiphiles for Efficient Intracellular and In Vivo Protein Delivery

Jingke Xu et al.

Summary: This research proposes a strategy for efficient intracellular protein delivery using dynamic polymer supra-amphiphiles, which promote the protein delivery process through the formation of dynamic supramolecular structures, showing great potential in treating diseases such as ulcerative colitis in vivo.

Real-Time Luminescence Assay for Cytoplasmic Cargo Delivery of Extracellular Vesicles

Masaharu Somiya et al.

Summary: This study developed an EV cargo delivery assay using a NanoBiT system, enabling real-time quantitative measurement of EV cargo delivery efficiency. It was found that modification of EVs with a virus-derived fusogenic protein significantly enhanced cytoplasmic cargo delivery.

Quantification of protein cargo loading into engineered extracellular vesicles at single-vesicle and single-molecule resolution

Andreia M. Silva et al.

Summary: This study introduced a workflow for accurately quantifying the efficiency of different EV-sorting proteins in promoting cargo loading into EVs. By using a combination of techniques, TSPAN14, CD63, and CD63/CD81 fused to the PDGFR beta transmembrane domain were identified as the most efficient EV-sorting proteins, accumulating an average of 50-170 single GFP molecules per vesicle.
