Towards Standardisation of a Diffuse Midline Glioma Patient-Derived Xenograft Mouse Model Based on Suspension Matrices for Preclinical Research

Summary: Diffuse midline glioma (DMG) is an aggressive brain tumor with limited treatment options. This study evaluated the HSJD-DIPG-007 patient-derived xenograft (PDX) model with respect to its ability to mimic human disease progression for therapeutic testing in vivo. The results showed that using Matrigel as a suspension matrix is a reliable method for establishing a DMG PDX mouse model, with delayed metastases formation, and is a step towards obtaining a standardized in vivo PDX model.

Inhibitors of PARP: Number crunching and structure gazing

Summary: Selective inhibitors of PARP1 and PARP2 are used for treating cancer patients with DNA repair deficiencies. Talazoparib shows higher potency compared to other inhibitors due to its extensive interactions with conserved residues in the active site. The recently characterized protein HPF1 may play a role in designing the next generation PARP1/2 inhibitors.

This review identifies and summarizes the parameters that influence the efficacy and safety outcomes of this approach, including microbubbles, transducer frequency, peak-negative pressure, pulse characteristics, and ultrasound dosing. Greater standardization of protocols and parameters is needed for robust clinical translation.

PARP Inhibitors for Breast Cancer: Germline BRCA1/2 and Beyond

Summary: PARP inhibitors are effective against breast cancers with mutations in DNA repair genes, especially BRCA1 and BRCA2. Olaparib and talazoparib are two FDA-approved PARPi for breast cancer treatment. Combination of Olaparib and Radiation Therapy for Triple Negative Breast Cancer: Preliminary Results of the RADIOPARP Phase 1 Trial

Summary: Preclinical studies have shown that TNBC cell lines are more sensitive to PARP inhibitors, leading to the development of new therapeutic approaches. The RADIOPARP phase 1 trial aimed to evaluate the toxicity and maximum tolerated dose of olaparib in combination with radiation therapy for TNBC patients. The trial showed that olaparib at a dose of 200 mg twice a day was well tolerated without dose-limiting toxicities.

Radiosensitizing Pancreatic Cancer with PARP Inhibitor and Gemcitabine: An In Vivo and a Whole-Transcriptome Analysis after Proton or Photon Irradiation

Summary: The study evaluated the synergistic effect of veliparib with radiation and temozolomide in patients with MGMT-unmethylated glioblastoma. Summary: This study describes a detailed procedure for establishing an ex vivo test coculture system of pediatric tumor organoids and immune cells to evaluate the efficacy of immunotherapies. This new platform allows for real-time visualization, monitoring, and robust quantification of tumor organoid cell cytotoxicity. The feasibility and sensitivity of this ex vivo coculture system using human immune cells and pediatric tumor organoids as ex vivo tumor models have been demonstrated.
