Summary: Jugulotympanic HNPGLs have distinct features, primarily occurring in women and having a lower proportion of SDHx mutations. Compared to carotid body and vagal HNPGLs, jugulotympanic HNPGLs are less likely to be bilateral, smaller in size, and have a lower metastatic rate. Patients with HNPGLs have higher plasma concentrations of MTY, while plasma normetanephrine does not differ significantly.

Quantifying evidence toward pathogenicity for rare phenotypes: The case of succinate dehydrogenase genes, SDHB and SDHD

Summary: The weight of evidence for the observation of rare missense variants in SDHB or SDHD in individuals with rare neuroendocrine tumors is uncertain. This study compared the frequency of these variants in cases of pheochromocytomas and paragangliomas (PCC/PGL) with population controls and calculated likelihood ratios for their pathogenicity. Regional enrichments of these variants and subphenotypic likelihood ratios were also measured. The study found that these rare missense variants can provide substantial evidence toward pathogenicity in PCC/PGL.

Serum fatty acid profiling in patients with SDHx mutations: New advances on cellular metabolism in SDH deficiency

Summary: Apart from succinate, little research has been done on extra-mitochondrial pathways in Succinate Dehydrogenase (SDH) genetic deficiency. Recent studies have highlighted the role of NADH/NAD(+) redox status and dependent pathways. In this study, FA metabolism data were collected from 30 patients with SDHx gene loss of function variants and 22 wild-type SDHx controls. The findings suggest changes in fatty acid metabolism, particularly in saturated FA elongation, shortening/elongation balance, and desaturation rates of C20-C24 PUFAs, in SDH-deficient patients with pheochromocytoma/paraganglioma (PPGL).

Succinate: A Serum Biomarker of SDHB-Mutated Paragangliomas and Pheochromocytomas
