Randomized clinical trial: effect of low-dose flutamide on abdominal adipogenic function in normal-weight women with polycystic ovary syndrome

Daniel A. Dumesic et al.

Summary: This study aimed to examine the effects of low-dose flutamide administration on abdominal fat deposition, lipid accumulation in adipocytes derived from stem cells, and glucose-lipid metabolism in normal-weight women with PCOS. The results showed that flutamide reduced abdominal fat deposition, attenuated lipid accumulation in adipocytes, and altered glucose-lipid homeostasis in PCOS women.

The mitochondrial profile in women with polycystic ovary syndrome: impact of exercise

Melpomeni Malamouli et al.

Summary: Polycystic ovary syndrome (PCOS) is a common endocrine disorder in pre-menopausal women with metabolic dysregulation. While exercise, especially high-intensity interval training (HIIT), is recommended as the first-line therapy for PCOS, the understanding of mitochondrial characteristics and their role in PCOS pathogenesis is limited. Further comprehensive studies are needed to investigate the association between metabolic dysfunction and aberrant mitochondrial profile, and to understand the molecular mechanisms underlying exercise-induced metabolic adaptations in women with PCOS.

Summary: The study estimated the excess prevalence and economic burden of pregnancy-related and long-term health morbidities attributable to PCOS, showing an additional total healthcare-related economic burden of $4.3 billion annually in the United States. Together with prior analysis, the economic burden of PCOS is estimated at $8 billion annually in 2020 USD.

The Complex Roles of Adipokines in Polycystic Ovary Syndrome and Endometriosis

Susanne Schueler-Toprak et al.

Summary: This review article provides an update on the role of adipokines in polycystic ovary syndrome (PCOS) and endometriosis, as well as summarizes previous findings. The article also discusses the involvement of single nucleotide polymorphisms (SNPs) in genes coding for adipokines and their receptors, the secretome of adipocytes, and epigenetic alterations of adipokine genes. Furthermore, the article addresses the notable effects of brown adipose tissue (BAT) on PCOS.

Cellular and Molecular Mechanisms of Metformin Action

Traci E. LaMoia et al.

Summary: Metformin is a commonly used first-line therapy for type 2 diabetes, mainly acting through inhibition of hepatic gluconeogenesis to lower blood glucose levels. Despite controversy over its mechanism of action, recent studies have supported a redox-dependent mechanism.

Accelerated subcutaneous abdominal stem cell adipogenesis predicts insulin sensitivity in normal-weight women with polycystic ovary syndrome

Daniel A. Dumesic et al.

Summary: In vitro differentiation of adipose stem cells into adipocytes in SC abdominal fat may predict insulin sensitivity in vivo in normal-weight women with PCOS, suggesting a potential role for hyperandrogenism in metabolic thrift. PCOS patients exhibited higher androgen levels, adipose-IR, and android fat percentage, while their adipocyte differentiation and gene expression in vitro were negatively correlated with adipose-IR and serum free testosterone but positively correlated with insulin sensitivity.

Menstrual dysfunction in polycystic ovary syndrome: association with dynamic state insulin resistance rather than hyperandrogenism

Summary: In women with PCOS, those with menstrual cycle abnormalities have higher levels of insulin resistance (IR) but not hyperandrogenism (HA), suggesting that IR, and not HA, plays a role in determining the degree of menstrual dysfunction and can be used as a clinical marker for IR in PCOS.

Absent Exercise-Induced Improvements in Fat Oxidation in Women With Polycystic Ovary Syndrome After High-Intensity Interval Training

Sofie Lionett et al.

Summary: The study found that women with PCOS did not show improvements in whole-body fat oxidation during submaximal exercise and mitochondrial respiration after 16 weeks of high-intensity interval training, despite an increase in peak oxygen uptake. In contrast, non-PCOS women demonstrated improvements in whole-body fat oxidation post high-intensity interval training, suggesting metabolic inflexibility in women with PCOS.
