Interrogation of the mammalian gut brain axis using LC-MS/MS-based targeted metabolomics with in vitro bacterial and organoid cultures and in vivo gnotobiotic mouse models

Thomas D. Horvath et al.

Summary: This article presents a quantitative analytical platform for studying the gut-brain axis, including the analysis of signals generated by microbes and hosts, as well as the preparation of gut-brain axis model systems. The platform combines microbial and organoid cultures with in vivo samples and uses liquid chromatography-tandem mass spectrometry-based metabolomics methods for analyzing microbially derived metabolites. The incorporation of three experimental models enhances the impact of this protocol. This article is of high importance.

Anthracycline-induced cardiotoxicity-are we about to clear this hurdle?

Wolfram C. M. Dempke et al.

Summary: Anthracyclines are highly effective cytostatic drugs used in cancer treatment, but they also cause significant cardiotoxicity. The development of reactive oxygen species and inhibition of topoisomerase II beta are believed to be the key mechanisms responsible for cardiotoxicity. To prevent this, various strategies, including drug therapy and the development of non-cardiotoxic analogues, are being pursued.

Suppression of high-fat-diet-induced obesity in mice by dietary folic acid supplementation is linked to changes in gut microbiota

Si Chen et al.

Summary: This study found that dietary folic acid supplementation inhibited weight gain and regulated gene expression related to fat and energy metabolism in obese mice by affecting gut microbiota. However, folic acid supplementation had almost no effect on weight and gene expression in germ-free mice.

Unravelling Mechanisms of Doxorubicin-Induced Toxicity in 3D Human Intestinal Organoids

Daniela Rodrigues et al.

Summary: This study investigates the molecular mechanisms associated with doxorubicin-induced intestinal toxicity using 3D small intestine and colon organoids. The study found that doxorubicin affects cell cycle, p53 signaling pathway, and oxidative stress in both types of organoids, with significant differences between colon and small intestine organoids in essential metabolic pathways. The study provides new insight into doxorubicin-induced effects on gene expression levels in the intestines.

Dietary Vitamin B1 Intake Influences Gut Microbial Community and the Consequent Production of Short-Chain Fatty Acids

Jonguk Park et al.

Summary: This study reveals the significant effect of vitamin B1 on the survival and competition of gut bacteria, providing a solution for individual differences in the gut microbial community. Additionally, vitamin B1 is involved in the production of butyrate. Through animal experiments and pathway analysis, possible associations and mechanistic insights are obtained.

Evaluation of hybrid surface technology for the analysis of the B-group vitamins by LC-ESI-MS/MS

Jinchuan Yang et al.

Summary: The novel hybrid surface technology (HST) has been developed to reduce metal analyte adsorption in liquid chromatography. The HST system showed significant improvements in sensitivity, LOQ, carry-over, and peak shape, particularly for B group vitamins such as riboflavin, thiamine, nicotinamide, FMN, PLP, and 5MTHF. The HST system demonstrated excellent accuracy and repeatability when applied to the analysis of B group vitamins in energy drinks and B vitamin dietary supplement samples.

B Vitamins and Their Roles in Gut Health

Khandkar Shaharina Hossain et al.

Summary: B vitamins play crucial roles as coenzymes in various cellular reactions, such as energy production and neurotransmitter synthesis. Recent studies have revealed novel functions of B vitamins in gut microbial ecology and intestinal health. This review highlights the functional roles, biosynthesis, and effects of B vitamins on gut microbiota growth and proliferation, as well as their impact on intestinal morphology, inflammation, and disorders.

Intestinal Microbiota Influence Doxorubicin Responsiveness in Triple-Negative Breast Cancer

Alaa Bawaneh et al.

Summary: This study demonstrates the impact of neoadjuvant chemotherapy on gut microbiota composition in triple-negative breast cancer (TNBC) and suggests that gut microbiota could be used as a predictive biomarker for doxorubicin responsiveness. Modulating gut microbiota through antibiotics, diet-derived fecal microbiota transplantation, or by exogenous LPS administration can influence tumor growth, treatment response, and metastasis formation.
