The Role of Tryptophan Metabolism in Alzheimer's Disease

Karl Savonije et al.

Summary: The search for new biochemical mechanisms for treating Alzheimer's disease is essential due to the limitations of current amyloid-based approaches. Amino acid metabolism, specifically tryptophan metabolism, is a potential alternative biological target. Tryptophan and its metabolites can inhibit enzymes involved in beta-amyloid synthesis and directly inhibit neurotoxic beta-amyloid oligomerization. Additionally, tryptophan metabolites can affect microglia and associated cytokines, contributing to neuroinflammation in Alzheimer's disease. The involvement of tryptophan biochemistry in Alzheimer's disease offers multiple druggable targets in the quest for therapeutics.

Loss of fatty acid degradation by astrocytic mitochondria triggers neuroinflammation and neurodegeneration

Yashi Mi et al.

Summary: Mi, Qi et al. have identified a mechanism in which defective oxidative phosphorylation in astrocytes disrupts astroglial lipid homeostasis, leading to neuronal damage and microglial reactivity. This mechanism involves the accumulation of lipid droplets in astrocytes, which triggers neurodegeneration resembling Alzheimer's disease. The study reveals the importance of astrocytic oxidative phosphorylation in fatty acid degradation and lipid homeostasis maintenance, and highlights the role of astrocyte dysfunction in neuroinflammation and neurodegeneration.

Integrative metabolomics-genomics approach reveals key metabolic pathways and regulators of Alzheimer's disease

Emrin Horgusluoglu et al.

Summary: This study aims to identify AD-specific metabolomic changes and their potential upstream genetic and transcriptional regulators through an integrative systems biology framework. The findings suggest that short-chain acylcarnitines/amino acids and medium/long-chain acylcarnitines are most associated with AD clinical outcomes. Integration of gene expression data reveals ABCA1 and CPT1A are involved in regulating acylcarnitines and amino acids in AD.

Kynurenine Metabolism and Alzheimer's Disease: The Potential Targets and Approaches

Vivek Kumar Sharma et al.

Summary: l-tryptophan, an essential amino acid, plays a role in regulating protein homeostasis and neurotransmitter-mediated physiological events. The metabolism of tryptophan through the kynurenine route leads to the production of metabolites that influence various physiological and pathological outcomes. Imbalances in the kynurenine pathway have been associated with several neurological disorders, including Alzheimer's disease. Pharmacological modulation of this pathway shows promise as a potential therapy for Alzheimer's disease.

Higher Serum DHA and Slower Cognitive Decline in Patients with Alzheimer's Disease: Two-Year Follow-Up

Che-Sheng Chu et al.

Summary: This study investigated the association between omega-3 polyunsaturated fatty acids (PUFAs) and cognitive function in patients with Alzheimer's disease (AD) receiving acetylcholinesterase inhibitors (AChEIs). The study found that lower levels of omega-3 PUFAs were associated with a higher risk of cognitive decline in AD patients. Lower baseline levels of docosahexaenoic acid (DHA), but not eicosapentaenoic acid (EPA), were also associated with a higher risk of cognitive decline. Higher scores on the Mini-Nutritional Assessment and total fat intake were independently associated with slower cognitive decline in AD patients receiving AChEIs.

Lipid accumulation induced by APOE4 impairs microglial surveillance of neuronal-network activity

Matheus B. Victor et al.

Summary: APOE4 induces a lipid-accumulated state in microglia, making them weakly responsive to neuronal activity. Neuronal cues differentially induce a lipogenic program in APOE4 microglia, exacerbating pro-inflammatory signals. APOE4 microglia disrupts the coordinated activity of neuronal ensembles through decreased uptake of extracellular fatty acids and lipoproteins.

THE COMPLEXITY OF ALZHEIMER'SDISEASE: AN EVOLVING PUZZLE

Camilla Ferrari et al.

Summary: The history of Alzheimer's disease dates back to 1907, but it wasn't until the end of the century that the components of pathological hallmarks and genetic subtypes were identified, leading to the first pathogenic hypothesis. Advances in biomarkers and technology have shifted the concept of AD from a static view to a biological entity that can manifest in various ways. Research shows that AD is heterogeneous in terms of amyloid composition, tau distribution, clinical symptoms, and genetic background, making it difficult to explain with a single pathological process.
