Infrared Diffusion-Ordered Spectroscopy Reveals Molecular Size and Structure

Summary: Inspired by NMR, IR-DOSY is a technique that characterizes both molecular structure and size using infrared spectroscopy. It achieves sensitivity to the diffusion coefficient by creating a concentration gradient and tracking its equilibration. Summary: Soluble oligomers of the amyloid-beta(1-42) peptide, which are considered neurotoxic in Alzheimer's disease, were characterized using biochemical and biophysical methods. The size of the oligomers was found to correlate with the position of the main amide I' band in the IR spectra, with larger oligomers having lower wavenumbers. Time-resolved aggregation experiments showed a more homogeneous absorption of beta-sheets in the oligomers after several hours, due to the consumption of smaller oligomers.

Detection of Glycine as a Model Protein in Blood Serum Using 2D-IR Spectroscopy

By combining data acquisition schemes and peak pattern analysis, the spectral signature of Gly can be separated from dominant protein fractions, with a detection limit of around 3 mg/mL. The potential for 2D-IR to complement IR absorption methods for measuring concentrations of amino acids, peptides, and low-molecular-weight proteins in serum samples is highlighted.

Infrared thermospectroscopic imaging of heat and mass transfers in laminar microfluidic reactive flows
