Protein conformational changes and protonation dynamics probed by a single shot using quantum-cascade-laser-based IR spectroscopy

Summary: Mid-IR spectroscopy is used to investigate protein reactions. In this study, quantum-cascade-laser-based dual-comb spectroscopy is used to probe protein conformational changes and protonation events by a single-shot experiment. The feasibility of QCL-based infrared spectroscopy in monitoring irreversible reactions is demonstrated.

Summary: The research focuses on the role of glutamines in the early stages of polyQ fibril formation, designing various template-assisted monomers with glutamine-rich sequences to study their effects on structural dynamics. Results show that even a few glutamines can destabilize the structure, and increasing the number of interacting glutamines and concentration are key factors for oligomerization. Time-resolved IR measurements indicate a slower kinetic phase upon oligomer formation.
