Tuberculosis drug discovery: Progression and future interventions in the wake of emerging resistance

Summary: The emergence of drug-resistant strains of tuberculosis poses a global health concern. Treatment of multidrug-resistant/extensively drug-resistant TB is complicated and requires second-line drugs with greater side effects. Recent approvals of new drugs for MDR/XDR-TB have provided impetus for TB drug discovery, highlighting the importance of combating TB through multidisciplinary approaches and leveraging recent advances in understanding the disease.

Identification of Potent DNA Gyrase Inhibitors Active against Mycobacterium tuberculosis

Summary: This study identified two potential inhibitors of M. tuberculosis DNA gyrase ATPase activity through virtual screening and subsequent biological assays. These compounds showed good antitubercular activity and likely exert their effects by binding to DNA gyrase. Molecular dynamics simulations and pharmacophore mapping analysis revealed the interaction mechanism of these compounds with the target enzyme, providing valuable insights for optimizing antitubercular agents.
