Summary: Expression of fission yeast Pho1 acid phosphatase is repressed by transcription of an upstream prt lncRNA and regulated by inositol pyrophosphate kinase Asp1. The 14-3-3 protein Rad24 is identified as a regulator of phosphate homeostasis. Mutations in Rad24 lead to increased Pho1 activity, and epistasis analysis reveals specific factors involved in this regulation. This study uncovers a new role for 14-3-3 protein as an inhibitor of RNA processing.

Genome-wide screens in yeast models towards understanding chronological lifespan regulation

Summary: Cellular models such as yeasts, which have simpler genomes, shorter lifespans and abundant genetic resources, play an important role in biogerontology research. Yeasts have contributed to the understanding of fundamental aspects of lifespan regulation, including nutrient response, protein translation, DNA damage, oxidative stress, mitochondrial function and autophagy. The use of automation and computational tools has allowed for the manipulation and analysis of genome-wide lifespan data, leading to a comprehensive view of cellular ageing.

Summary: We studied the biochemical properties and mutations of the autonomous Asp1 kinase domain in fission yeast. The results showed that under optimal kinase conditions, the reaction of Asp1 kinase with IP6 and ATP resulted in both IP6 phosphorylation to 1-IP7 and hydrolysis of the ATP gamma-phosphate with near-equal partitioning between productive 1-IP7 synthesis and unproductive ATP hydrolysis.

Sporulation: A response to starvation in the fission yeast Schizosaccharomyces pombe

Response to leucine in Schizosaccharomyces pombe (fission yeast)

Summary: Leucine (Leu) is an essential branched-chain amino acid in animals, including humans. Although Leu auxotrophy is inconvenient for the fission yeast Schizosaccharomyces pombe, it has enhanced its usefulness as a model organism in laboratories. Leu auxotrophy induces intracellular responses and phenotypes different from those of the prototrophic strains, which necessitates caution in the analysis and interpretation of experimental results. Understanding the intracellular responses caused by Leu auxotrophy can contribute to the research using this model organism and to the understanding of intracellular responses in higher organisms. This minireview summarizes the latest research on leucine metabolism mechanism and starvation response in the fission yeast Schizosaccharomyces, along with the identification of the ilv3+ gene involved in leucine synthesis.

Screening of selected ageing-related proteins that extend chronological life span in yeast Saccharomyces cerevisiae

Summary: Proteins that extend yeast chronological life span were identified through screening of ageing-related proteins. Three of these proteins, Ptc4, Zwf1, and Sme1, showed enhanced survival percentage and shorter doubling time of yeast growth compared to control. These proteins contributed differently to thermal and oxidative stress responses, suggesting distinct mechanisms of actions. The overexpression of Ptc4 or Zwf1 also promoted rapid cell proliferation during yeast growth, indicating their involvement in cell division or growth pathways.

Regulation of inorganic polyphosphate is required for proper vacuolar proteolysis in fission yeast

Summary: The SPX-RING-type ubiquitin ligase Pqr1 is crucial for maintaining quiescence in Schizosaccharomyces pombe by regulating proper levels of phosphate and its polymer polyphosphate, which in turn affect vacuolar proteolysis and autophagy. Deletion of Pqr1 leads to excessive accumulation of intracellular phosphate and polyphosphate, disrupting autophagy-dependent proteolysis under nitrogen starvation. These findings highlight the importance of polyphosphate metabolism and vacuolar function in maintaining cellular viability during quiescence.

This regulation involved transcription factor COH2 activating expression of cuticle penetration genes on insect cuticle, and protein COH1 interacting with COH2 to down-regulate cuticle penetration genes and up-regulate genes for hemocoel colonization in the hemocoel. These findings provide valuable insights into fungal pathogenicity in insects.
