Association of In Utero Antipsychotic Medication Exposure With Risk of Congenital Malformations in Nordic Countries and the US

Summary: In this study, in utero antipsychotic exposure was generally not significantly associated with an increased risk of malformations, although increased risks were observed for specific combinations. The association between olanzapine and oral clefts, gastroschisis and other specific brain anomalies with atypical antipsychotics, and cardiac malformations with chlorprothixene should be further investigated.

Increasing Prevalence and High Risk of Associated Anomalies in Congenital Vertebral Defects: A Population-based Study

Maternal risk factors for congenital limb deficiencies: A population-based case-control study

Summary: This nationwide population-based case-control study in Finland found that pregestational diabetes, primiparity, and young maternal age are associated with an increased risk of congenital limb deficiencies. Pregnant women taking medications, especially antiepileptics, or with pregestational diabetes should be carefully monitored for the occurrence and risk of limb deficiencies in the fetus.

Prenatal exposure to antipsychotic agents and the risk of congenital malformations in children: A systematic review and meta-analysis

Summary: Thirteen studies were included in the systematic review, with six studies included in the meta-analysis, showing a pooled adjusted risk ratio of 1.23 for the association between prenatal exposure to antipsychotic medications and the risk of congenital malformations in children. Further studies focusing on specific medication classes and dose responses are recommended to assist clinicians in making prescribing decisions during pregnancy.
