The anticancer and EGFR-TK/CDK-9 dual inhibitory potentials of new synthetic pyranopyrazole and pyrazolone derivatives: X-ray crystallography, in vitro, and in silico mechanistic investigations

Summary: The inadequate treatment options for breast cancer are due to the lack of targeted medications, leading to the recurrence of metastatic disorders. Overexpression of EGFR-TK and CDK-9 in breast cancer patients is associated with worse overall survival. We synthesized two series of suggested anticancer molecules and characterized them using various methods. Unexpectedly, our attempt to obtain pyranopyrazole-gold complexes yielded benzylidene-2,4-dihydro-3H-pyrazol-3-one instead. The synthesized compounds showed variable cytotoxicity and effectively inhibited EGFR-TK and CDK-9.

Exploring the dual effect of novel 1,4-diarylpyranopyrazoles as antiviral and anti-inflammatory for the management of SARS-CoV-2 and associated inflammatory symptoms

Summary: This study reports the design and synthesis of a series of sixteen pyranopyrazole derivatives substituted with two aryl groups. The designed compounds are suggested to show dual activity against emerging Coronaviruses and associated inflammations. Results reveal that three pyranopyrazoles have the potential to inhibit the viral main protease and the most active compound also demonstrates anti-inflammatory effects. Further evaluation and optimization of these compounds are recommended for COVID-19 treatment.

Novel benzenesulfonamide-bearing pyrazoles and 1,2,4-thiadiazoles as selective carbonic anhydrase inhibitors

Summary: Two series of novel benzenesulfonamides were synthesized and tested as hCA inhibitors, with amino-linked 1,2,4-thiadiazoles showing better inhibition of hCA I compared to thioureido-linked pyrazoles. Most of the newly synthesized sulfonamides exhibited strong inhibition of hCA II, while compound 8e was found to be the most selective inhibitor of hCA II.

Unravelling the antifungal and antiprotozoal activities and LC-MS/MS quantification of steroidal saponins isolated from Panicum turgidum

Summary: Bioassay-guided investigation of Panicum turgidum extract led to the discovery of seven steroidal saponins (Turgidosterones 1-7), which showed antifungal, antileishmaniat, and antitrypanosomal activities. The most active antifungal compound was Turgidosterone 6, while Turgidosterones 4-7 exhibited significant activities against Leishmania donovani and Trypanosoma brucei. The structural features of spirostane-type saponins and the elongation of sugar residue at C-3 were found to affect the activities. Additionally, a LC-MS/MS method was used to quantitatively determine the abundance of active ingredients in Panicum turgidum extract.

Robust antiviral activity of commonly prescribed antidepressants against emerging coronaviruses: in vitro and in silico drug repurposing studies

The effect of novel synthetic semicarbazone- and thiosemicarbazone-linked 1,2,3-triazoles on the apoptotic markers, VEGFR-2, and cell cycle of myeloid leukemia

Summary: VEGFR-2 is a rational target in cancer therapy. Current VEGFR-2 inhibitors have potent antitumor activity but also serious unwanted effects. In this study, a new set of 1,2,3-triazole derivatives were synthesized to inhibit VEGFR-2 signaling. These compounds showed growth inhibition effect against a variety of cancer cells, with thiosemicarbazone-linked compounds being the most active. Further experiments revealed that one compound could induce apoptosis and cell cycle arrest. Molecular docking studies confirmed the binding potential of these compounds to VEGFR-2.

In vitro and computational investigations of novel synthetic carboxamide-linked pyridopyrrolopyrimidines with potent activity as SARS-CoV-2-MPro inhibitors

Summary: A novel set of compounds was designed and evaluated for their antiviral activity against COVID-19 using in vitro assays and molecular docking simulations. The majority of these compounds showed potent antiviral activity with minimal cytotoxicity. Compounds 25 and 29 exhibited promising inhibitory activity and were stable in molecular dynamic simulations. These results suggest that compounds 25 and 29 are potential drug candidates for the treatment of SARS-CoV-2.
