The amyloid concentric beta-barrel hypothesis: Models of amyloid beta 42 oligomers and annular protofibrils

Stewart R. Durell et al.

Summary: Amyloid beta (A beta) peptides, particularly A beta 42, contribute to Alzheimer's disease. Molecular models of A beta 42 hexamers have been proposed, and experimental findings support various aspects of these models. Recent studies have also proposed structures for A beta 42 tetramers and octamers. Differing assembly types for A beta 42 have been identified, and concentric beta-barrel models have been proposed.

Beta-amyloid pore linked to controlled calcium influx into the cell: A new paradigm for Alzheimer's Disease

Martina Pannuzzo

Summary: Despite continuous worldwide efforts, clinical trials for Alzheimer's disease (AD) therapeutics have not been successful. There is a new concept suggesting that the amyloid-beta (Aβ) peptide may modulate synaptic plasticity and protect against excitotoxicity, challenging the previous belief that it is a major contributor to AD pathogenesis. The relationship between Aβ-mediated synaptic plasticity and trafficking remains complex and controversial, indicating a need for further research.

Molecular Dynamics Simulations Based on Polarizable Models Show that Ion Permeation Interconverts between Different Mechanisms as a Function of Membrane Thickness

Peiran Chen et al.

Summary: The study used molecular dynamics simulations to characterize the permeation process of charged compounds through lipid membranes of different thicknesses. It was found that in thin membranes, ion translocation is primarily through an ion-induced defect mechanism, while in thicker membranes it transitions to a solubility-diffusion mechanism. This highlights the importance of induced polarizability in accurately modeling the permeation of charged molecules through lipid membranes.

Summary: Protein misfolding and aggregation are common in amyloidogenic diseases, and understanding how amyloid species become toxic is crucial for treatment development. Experiments using computer modeling, in vitro and in vivo studies, as well as pharmacology, have provided valuable insights into the accumulation and deposition of amyloid species, contributing to research on diseases like Alzheimer's, Parkinson's, diabetes, and ALS.

Cryo-electron Microscopy Imaging of Alzheimer's Amyloid-beta 42 Oligomer Displayed on a Functionally and Structurally Relevant Scaffold

Jinming Wu et al.

Summary: Amyloid-beta peptide (Aβ) oligomers are pathogenic aggregates in Alzheimer's disease, which can permeabilize cellular membranes through a pore formation mechanism. The Aβ42-αHL oligomers share major structural, functional, and biological properties with wild-type Aβ42-pores, with the pore region being intrinsically flexible according to single-particle cryo-EM analysis. Aβ42-αHL oligomers could be a highly specific antigen for immuno-based diagnostics and therapies, allowing for the study of features of wild-type amyloid oligomers.
