Ion-Ion Charge Reduction Addresses Multiple Challenges Common to Denaturing Intact Mass Analysis

Summary: Complete LC-MS-based protein primary sequence characterization requires measurement of intact protein profiles under denaturing and/or reducing conditions. By using broadband isolation of entire charge state distributions of intact proteins followed by ion-ion proton transfer charge reduction, the method proposed in this study, termed full scan PTCR (fsPTCR), addresses issues of protein overcharging and sample heterogeneity, leading to significant analytical advantages.

Streamlining the Characterization of Disulfide Bond Shuffling and Protein Degradation in IgG1 Biopharmaceuticals Under Native and Stressed Conditions

Summary: This study investigates the effects of shuffled disulfide bonds on the safety and efficacy of protein therapeutics by comparing two IgG1 protein therapeutics, rituximab and bevacizumab. The results show differences in the degradation of the two proteins, with bevacizumab exhibiting an increase in shuffled disulfide bond levels over time while rituximab maintains similar levels. However, there were no significant variations between the originator and biosimilar versions of the drugs in terms of disulfide shuffling and overall degradation. This study provides important insights for the development and regulatory approval of biosimilars.
