The mitochondrial redistribution of eNOS is involved in lipopolysaccharide induced inflammasome activation during acute lung injury

Hui Wang et al.

Summary: Acute lung injury (ALI) is a devastating clinical syndrome with no effective therapies, where inflammasome activation plays a critical role. The redistribution of uncoupled eNOS in mitochondria could be a key factor in activating the inflammatory response in ALI. Inhibiting mitochondrial reactive oxygen species (mito-ROS) generation may be a potential therapeutic strategy for ALI.

Function and regulation of the divisome for mitochondrial fission

Felix Kraus et al.

Summary: Mitochondria form dynamic networks through fusion and fission events, with fission serving as an endpoint event in signaling axis for cells to sense and respond to external cues. The fission process is coordinated by membrane-associated adaptors, influenced by organellar and cytoskeletal interactions, and executed by the GTPase DRP1. Separating the functional and regulatory aspects of the 'mitochondrial divisome' into core and accessory machinery provides a mechanistic understanding of the mitochondrial fission process.

PINK1-induced phosphorylation of mitofusin 2 at serine 442 causes its proteasomal degradation and promotes cell proliferation in lung cancer and pulmonary arterial hypertension

Asish Dasgupta et al.

Summary: The study identified that excessive phosphorylation of Mfn2 leads to proteasomal degradation in PAH and NSCLC, and inhibiting this phosphorylation may hold therapeutic potential for these diseases. By modifying Mfn2 constructs, it was shown that increasing Mfn2 expression can suppress cell proliferation and induce apoptosis in these conditions.

TNF alpha induces mitochondrial fragmentation and biogenesis in human airway smooth muscle

Philippe Delmotte et al.

Summary: In hASM of asthmatic patients, mitochondrial volume density is higher and fragmentation is more common, likely influenced by proinflammatory cytokines. TNF alpha exposure leads to increased mitochondrial biogenesis, volume density, and fragmentation, ultimately reducing O-2 consumption rate per mitochondrion. These changes are associated with alterations in expression of key mitochondrial proteins and increased cell proliferation in hASM.
