Global burden of bacterial antimicrobial resistance in 2019: a systematic analysis

Christopher J. L. Murray et al.

Summary: Antimicrobial resistance (AMR) poses a significant threat to global human health, and this study provides the most comprehensive estimates of AMR burden to date. By estimating deaths and disability-adjusted life-years (DALYs) associated with bacterial AMR in 204 countries and territories in 2019, the study highlights the impact of resistance and the leading pathogen-drug combinations contributing to it.

Studies on the Reactions of Biapenem with VIM Metallo β-Lactamases and the Serine β-Lactamase KPC-2

Anka Lucic et al.

Summary: This study investigated the reaction of the carbapenem biapenem with different subclasses of beta-lactamases. The results showed the formation of enamine and various types of imine products. The findings support the idea that prolonging the lifetime of beta-lactamase carbapenem complexes by optimizing the tautomerization process could lead to improved carbapenems.

Cephalosporin Prodrug Inhibitors Overcome Metallo-β-Lactamase Driven Antibiotic Resistance

Matthijs J. van Haren et al.

Summary: The increasing prevalence of metallo-beta-lactamase (MBL)-expressing bacteria is a worrying trend in antibiotic resistance. Studies have shown that zinc ions, which are essential for the hydrolytic activity of MBLs, can be targeted for inhibition. Researchers have developed a series of cephalosporin prodrugs that exhibit potent activity against MBLs and can act synergistically with specific antibiotics.

Crystallography and QM/MM Simulations Identify Preferential Binding of Hydrolyzed Carbapenem and Penem Antibiotics to the L1 Metallo-β-Lactamase in the Imine Form

Rebecca M. Twidale et al.

Summary: The widespread bacterial resistance to carbapenem antibiotics, due to carbapenem-hydrolyzing enzymes including metallo-beta-lactamases, is a growing global health concern. A combined approach utilizing crystallography and quantum mechanics/molecular mechanics simulations allowed for the assignment of tautomers in M beta L:hydrolyzed antibiotic complexes, providing insights into beta-lactam hydrolysis mechanisms. The relative stability of different hydrolyzed forms of (carba)penems in the L1 active site was established, identifying important interactions for stable complex formation and potentially aiding in inhibitor design for combating antibiotic resistance.

Faropenem reacts with serine and metallo-β-lactamases to give multiple products

Anka Lucic et al.

Summary: Penems have shown potential as antibacterials and beta-lactamase inhibitors, but their clinical use is limited compared to carbapenems. Faropenem, with a C-2 tetrahydrofuran ring, is resistant to some beta-lactamases. Studies on reactions of faropenem with beta-lactamases demonstrate different outcomes, suggesting further research is needed for optimizing the interactions between penems and beta-lactamases.

Summary: Antimicrobial resistance is a major problem in current practical medicine, with the spread of genes coding for resistance determinants among bacteria challenging the use of antibiotics. Metallo-beta-lactamases play a central role in antibiotic resistance, with their diverse characteristics explored in the chemical, biochemical, and microbiological aspects.

Novel Cephalosporin Conjugates Display Potent and Selective Inhibition of Imipenemase-Type Metallo-β-Lactamases

Kamaleddin H. M. E. Tehrani et al.

Summary: The novel cephalosporin prodrugs designed to inhibit metallo-beta-lactamases have shown potential, especially the thiomandelic acid conjugate (8). Conjugate 8 not only effectively inhibits IMP-type MBLs, but also significantly reduces the minimum inhibitory concentration of meropenem against IMP-producing bacteria. These prodrugs act as slowly turned-over substrates to inhibit IMP-type MBLs, with the phenyl and carboxyl moieties of crucial importance for potency.
