Effect of silibinin on ethanol- or acetaldehyde-induced damge of mouse primary hepatocytes in vitro

Summary: Silibinin does not protect hepatocytes from ethanol-induced injury, but significantly reduces acetaldehyde-induced hepatocyte injury by down-regulating apoptosis and up-regulating autophagy, as demonstrated by cell viability assays and enzyme leakage tests. The study also shows that acetaldehyde induces autophagy in hepatocytes, with autophagy playing a protective role against apoptosis in this context. Silibinin further enhances this protective effect by modulating the levels of caspase-3 and LC3, suggesting a potential therapeutic mechanism for silibinin in preventing acetaldehyde-related liver damage.

Despite its ability to modulate a wide array of cellular responses, the major obstacle in declaring Silibinin as a translational chemotherapeutic agent lies in its lower bioavailability.
