Changing perspectives on frontotemporal dementia: A review

Summary: This article examines the evolution in understanding of frontotemporal dementia (FTD) during the last four decades. It discusses the recognition of heterogeneity in FTD, as it is now known to be associated with alterations in language, conceptual knowledge, and praxis, in addition to behavior and executive impairment. The article also explores the relationship between FTD and other neurological disorders, such as motor neurone disease (MND), progressive supranuclear palsy (PSP), and corticobasal degeneration (CBD). The underlying pathological proteins and genetic mutations in familial FTD are identified. The article emphasizes the importance of understanding the diverse nature of FTD for effective diagnosis, management, and the development of targeted therapies.

Progranulin as a therapeutic target in neurodegenerative diseases

Mutations in the GRN gene can cause neurodegenerative diseases, while mutations that reduce PGRN levels increase the risk of other neurological disorders. Genetic Spectrum and Clinical Heterogeneity of Chinese Frontotemporal Dementia Patients: Data from PUMCH Dementia Cohort

Involvement of striatal motoric subregions in familial frontotemporal dementia with parkinsonism harboring the C9orf72 repeat expansions

Summary: This study investigated the pathophysiological mechanism of parkinsonism caused by C9ORF72 repeat expansions in frontotemporal dementia with parkinsonism (FTDP). Clinical and imaging analysis of two FTDP patients and seventeen healthy controls revealed gray matter volume atrophy, hypometabolism, and dopamine dysfunction in the motor areas of the striatum. These findings suggest that C9ORF72 repeat expansions may contribute to the degeneration and dopaminergic dysfunction of the striatal motor subregion in FTDP patients with parkinsonism.

Autosomal Dominant Frontotemporal Lobar Degeneration in a Filipino Family with Progranulin Mutation

Summary: In this study, a Filipino family with familial frontotemporal lobar degeneration (FTLD) and a GRN gene mutation was presented, with 11 affected relatives across 3 generations. Most affected family members displayed behavioral variant frontotemporal dementia (FTD) and progressive nonfluent aphasia (PNFA). Neuroimaging revealed atrophy and hypometabolism consistent with FTD syndromes. The study highlights the importance of identifying affected families and cohorts for potential therapeutic developments in the field of FTLD-GRN.
