Psychiatric symptoms of frontotemporal dementia and subcortical (co-)pathology burden: new insights

Marta Scarioni et al.

Summary: The majority of patients with frontotemporal dementia have additional pathologies alongside the main pathology. Neuropsychiatric symptoms in FTD are associated with subcortical pathology in the hippocampus, with hallucinations linked to a higher burden of TDP-43 pathology in the granular layer. Different pathological proteins accumulate in various brain regions and contribute to the diverse clinical presentation of frontotemporal lobar degeneration (FTLD).

Altered reward processing underpins emotional apathy in dementia

Stephanie Wong et al.

Summary: This study found a significant link between impaired social reward learning and emotional apathy in dementia, suggesting a shared neurobiological basis between the two. Understanding the neurocognitive mechanisms of reward processing can help improve the identification and treatment of emotional apathy in dementia.

Examining empathy deficits across familial forms of frontotemporal dementia within the GENFI cohort

Empathy and Impaired Socioemotional Self-Perception in Frontotemporal Dementia

Mario F. Mendez et al.

Summary: Impaired empathy in behavioral variant frontotemporal dementia (bvFTD) may result from a lack of self-consciousness in social situations, according to this pilot study. These preliminary findings suggest that impaired socioemotional self-perception plays a role in the loss of empathy among patients with bvFTD.

Multidimensional Apathy in Behavioral Variant Frontotemporal Dementia, Primary Progressive Aphasia, and Alzheimer Disease

Ratko Radakovic et al.

Summary: The study examined apathy profiles and awareness in patients with behavioral variant frontotemporal dementia (bvFTD), primary progressive aphasia (PPA), and Alzheimer disease (AD), revealing emotional apathy as a prevalent feature in bvFTD with lower self-awareness, while lower self-awareness for executive apathy distinguishes bvFTD from PPA.

Emotion recognition of morphed facial expressions in presymptomatic and symptomatic frontotemporal dementia, and Alzheimer's dementia

Lize C. Jiskoot et al.

Summary: The study using the Emotion Recognition Task (ERT) revealed emotion recognition deficits in patients with behavioral variant FTD (bvFTD) and Alzheimer's Dementia (AD), while presymptomatic mutation carriers and controls performed relatively well. Results also showed that task performance varied with emotional intensity levels.

Evaluation of Discriminative Detection Abilities of Social Cognition Measures for the Diagnosis of the Behavioral Variant of Frontotemporal Dementia: a Systematic Review

Alessandra Dodich et al.

Summary: The use of social cognition measures in the diagnosis of bvFTD shows potential in clinical settings, especially in emotion recognition and theory of mind tasks. However, the lack of standardized cognitive measures, varied task formats, and limited normative data hinder study comparability. Further comparative studies are needed to determine the best choice for ensuring high diagnostic accuracy in bvFTD assessments.

The interplay of emotional and social conceptual processes during moral reasoning in frontotemporal dementia

Cherie Strikwerda-Brown et al.

Summary: The study found that patients with bvFTD could make judgments on different courses of action in moral scenarios, but their emotional responses to these decisions were irregular. This blunted emotional reaction was specific to personal high-conflict conditions and was correlated with reduced knowledge of socially acceptable behavior.

Progression of Behavioral Disturbances and Neuropsychiatric Symptoms in Patients With Genetic Frontotemporal Dementia

Alberto Benussi et al.

Summary: This cohort study uses data from tertiary frontotemporal dementia research clinics across Europe and Canada to assess the frequency and severity of behavioral symptoms and their progression in different forms of genetic frontotemporal dementia. Patients with MAPT variants showed the highest frequency and severity of most behavioral symptoms compared with C9orf72 and GRN carriers. Anxiety and depression were most common in GRN and MAPT carriers; hallucinations, particularly auditory and visual, were most common in C9orf72 carriers.

Uncovering the prevalence and neural substrates of anhedonia in frontotemporal dementia

Siobhan R. Shaw et al.

Summary: This study reveals profound anhedonia in frontotemporal dementia (FTD) syndromes, mainly caused by atrophy of frontostriatal brain regions specialized for hedonic tone, with distinct neural drivers compared to apathy or depression. Additional research is needed to understand the impact of anhedonia on daily activities and develop targeted interventions to improve quality of life in patients and their families.

Cortical [18F]PI-2620 Binding Differentiates Corticobasal Syndrome Subtypes

Carla Palleis et al.

Summary: The study showed that the tau PET ligand [F-18]PI-2620 has diagnostic value in patients with corticobasal syndrome, providing quantitatively and regionally distinct signals. Regardless of beta-amyloid status, the hemispheric lateralization of [F-18]PI-2620 signal reflects the severity of the disease clinically.

Plasma phosphorylated tau 217 and phosphorylated tau 181 as biomarkers in Alzheimer's disease and frontotemporal lobar degeneration: a retrospective diagnostic performance study

Elisabeth H. Thijssen et al.

Summary: Plasma tau phosphorylated at threonine 217 (p-tau217) and plasma tau phosphorylated at threonine 181 (p-tau181) showed excellent diagnostic performance in distinguishing patients with Alzheimer's disease syndromes from other neurodegenerative disorders. P-tau217 outperformed p-tau181 in differentiating Alzheimer's disease syndromes from FTLD syndromes, indicating amyloid-PET positivity, and showing stronger correlations with tau-PET signal.

Summary: This study investigated the difference in facial emotion recognition among Malaysian Malays and Australians with a European heritage with and without depression. Results showed that depression status and cultural group did not significantly influence overall FER accuracy. Malaysian participants without MDD and Australian participants with MDD performed quicker, and Malaysian participants more accurately recognized fear compared to Australian participants.

The role of neurofilament light chain in frontotemporal dementia: a meta-analysis

Eleni Karantali et al.

Summary: Neurofilament light chain (NfL) may play a role in differentiating patients with frontotemporal dementia from healthy controls, patients with Alzheimer's dementia, and psychiatric disorders. Additionally, higher levels of NfL were observed in patients with semantic primary progressive aphasia (PPA) compared to behavioral FTD and non-fluent PPA patients, suggesting the utility of NfL in distinguishing specific FTD subgroups. Further research exploring the use of NfL in frontotemporal dementia is warranted.
