The Alzheimer's Disease Mitochondrial Cascade Hypothesis: A Current Overview

Russell H. Swerdlow

Summary: Viable hypotheses about Alzheimer's disease must consider its age-dependence, commonality, association with specific biological factors, connection with various changes in the body, and systemic features. Mitochondria and factors influenced by mitochondria may link these different characteristics. The mitochondrial cascade hypothesis provides a straightforward explanation and accumulating data support its validity. While alternative hypotheses may also explain mitochondria-related phenomena, the primary mitochondrial cascade hypothesis will continue to evolve and attract interest.

Failure of DNA double-strand break repair by tau mediates Alzheimer's disease pathology in vitro

Megumi Asada-Utsugi et al.

Summary: Phosphorylated tau protein accumulates at DNA double-strand breaks (DSBs) in neurons, and failure of DSB repair leads to increased tau pathology, potentially associated with Alzheimer's disease.

Calcineurin dephosphorylates topoisomerase IIb and regulates the formation of neuronal-induced DNA breaks

Ilse Delint-Ramirez et al.

Summary: Stimulus-dependent calcium influx through NMDA receptors activates calcineurin phosphatase, which dephosphorylates Top2B and stimulates its DNA cleavage activity to generate DSBs. This process also occurs after physiological neuronal activity, highlighting the significance of calcineurin in regulating DSB formation.

DNA Double-Strand Break Accumulation in Alzheimer's Disease: Evidence from Experimental Models and Postmortem Human Brains

Nidheesh Thadathil et al.

Summary: Alzheimer's disease (AD) is characterized by progressive neurodegeneration with accumulation of DNA double-strand breaks (DSBs) playing an important role in its pathogenesis. Studies have shown increased DNA DSB accumulation and reduced repair function in the hippocampus of AD brains compared to non-AD brains. Additionally, altered levels of DNA repair proteins were observed in AD mouse models and cellular models, highlighting the potential influence of DNA DSB and repair proteins in the pathway toward neural damage and memory loss in AD.

DNA damage-how and why we age?

Matt Yousefzadeh et al.

Summary: Aging is a complex process that involves DNA damage leading to cells entering apoptosis or senescence to avoid replicating a damaged genome. DNA damage is also linked to other cellular processes implicated as drivers of aging, supporting the idea that DNA damage could be the root cause of aging.

APOE and Alzheimer's Disease: From Lipid Transport to Physiopathology and Therapeutics

Mohammed Amir Husain et al.

Summary: Alzheimer's disease is a devastating neurodegenerative disorder characterized by abnormal protein aggregations, with the APOE gene playing a role in the risk of developing the disease. ApoE is a major lipid transporter in the CNS, involved in development, maintenance, and repair, as well as regulating important signaling pathways. Various apoE-based therapeutics are being developed for the treatment of AD.

Single-nucleus chromatin accessibility and transcriptomic characterization of Alzheimer's disease

Samuel Morabito et al.

Summary: An integrative analysis of single-nucleus assay for transposase-accessible chromatin with sequencing and RNA sequencing in normal and Alzheimer's disease brain tissue reveals cell-type-specific cis-regulatory elements and candidate target genes. The study highlights the dynamic gene-regulatory landscape of the brain, identifying disease-related regulatory modules and transcription factors in specific cell types, demonstrating the utility of a multi-omic single-nucleus approach.
