Redox metabolism for improving whole-cell P450-catalysed terpenoid biosynthesis

Behnaz Nowrouzi et al.

Summary: The increasing trend towards producing cytochrome P450-mediated natural products in microbial systems is due to the challenging nature of organic chemistry methods. Manipulating the redox potential in microbial chassis for natural product synthesis is limited, as the crucial role of redox state in regulating P450 biocatalysis has been underestimated.

Astaxanthin Bioactivity Is Determined by Stereoisomer Composition and Extraction Method

Terry W. Snell et al.

Summary: Astaxanthin (ASX) is a powerful antioxidant that has significant effects on the metabolism of many animals, increasing reproductive capacity, egg yolk volume, growth rates, immune responses, and disease resistance. ASX targets mitochondria to protect them from damage by reactive oxygen species (ROS). Most animals cannot synthesize ASX and need to acquire it from their diet. However, ASX is easily denatured during extraction and its hydrophobicity limits its bioavailability.

Metabolic engineering of Escherichia coli with electron channelling for the production of natural products

Seon Young Park et al.

Summary: This study reports an electron channelling strategy based on the application of Photorhabdus luminescens CipB scaffold protein, which allows efficient electron transfer between eukaryotic cytochrome P450s (P450s) and P450 reductase in bacteria. Using this strategy, recombinant Escherichia coli strains producing various natural products were developed and the production titres were enhanced through optimization of the culture conditions.

An electron transfer competent structural ensemble of membrane-bound cytochrome P450 1A1 and cytochrome P450 oxidoreductase

Goutam Mukherjee et al.

Summary: Mukherjee, Nandekar and Wade investigate the structural arrangement of the complex between membrane-bound cytochrome P450 1A1 and NADPH-cytochrome P450 reductase. They find that upon binding to the reductase, the catalytic domain of cytochrome P450 1A1 reorients subject to the constraints of membrane binding, potentially explaining why the electron transfer rates between the proteins are low when compared to those of soluble bacterial cytochrome P450s.
