Imidazole-Based pH-Sensitive Convertible Liposomes for Anticancer Drug Delivery

Summary: In this study, three novel lipids were prepared and incorporated into PEGylated liposomes containing doxorubicin to enhance their activity against solid tumors. These lipids were designed to cluster with negatively charged phosphatidylethanolamine-polyethylene glycol at acidic tumor sites, thereby triggering the conversion of liposomes into pH-sensitive convertible liposomes. The pH drop resulted in increased positive surface charges, lipid phase separation, and aggregation with cell membrane-mimetic liposomes, leading to enhanced drug release and better anticancer activity against both monolayer cells and 3D multicellular spheroids.

Preparation and application of pH-responsive drug delivery systems

Summary: Microenvironment-responsive drug delivery systems (DDSs) achieve targeted drug delivery and improve drug efficacy by responding to specific pH conditions in diseased tissues. Various synthetic approaches have been developed to prepare pH-sensitive DDSs, including the introduction of pH-sensitive chemical bonds or groups. Different nano DDSs, such as liposomes, micelles, hydrogels, dendritic macromolecules, and organic-inorganic hybrid nanoparticles, can respond to pH variations. These pH-sensitive DDSs have been applied in research on anticancer therapy and the diagnosis of cancer, inflammation, antibacterial infection, and neurological diseases.

pH-Responsive PEGylated Niosomal Nanoparticles as an Active-Targeting Cyclophosphamide Delivery System for Gastric Cancer Therapy

Summary: In this study, a PEGylated niosomal formulation of cyclophosphamide was prepared and characterized. The prepared nanocarrier showed controlled drug release and improved stability. It induced apoptosis in cancer cells by regulating gene expression and inhibited cell cycle and migration. These results suggest that the developed nanocarrier could be an effective and targeted drug delivery system for gastric cancer therapy.

Impact of PEGylated Liposomal Doxorubicin and Carboplatin Combination on Glioblastoma

Summary: This study evaluated the therapeutic efficacy of doxorubicin and carboplatin loaded into PEG-modified liposome nanoparticles for glioblastoma treatment. The results showed that PEG-Lip-DOX/CB exhibited controlled release of the drugs and significantly increased their cytotoxicity against cancer cells. In vivo experiments also demonstrated that PEGylated liposomes improved therapeutic effects and reduced side effects compared to non-PEGylated liposomes.

New insights into the activities and toxicities of the old anticancer drug doxorubicin

Summary: Doxorubicin is a commonly used chemotherapeutic drug with limited application due to severe adverse effects like cardiotoxicity, therapy-related malignancies, and gonadotoxicity. Recent studies have shown that doxorubicin causes DNA double-strand breaks and chromatin damage, the latter playing a major role in drug efficacy. Efforts are being made to develop effective analogues with limited adverse effects by understanding the mechanisms of anthracycline activities and toxicities.
