Innovative nanotheranostics: Smart nanoparticles based approach to overcome breast cancer stem cells mediated chemo- and radioresistances

Summary: The increasing number of breast cancer cases globally and the rising death rate indicate the inadequacy of traditional and current treatments. Nanoparticle-based therapies such as photothermal therapy, photodynamic therapy, chemodynamic therapy, and sonodynamic therapy show promise in combating breast cancer through accurate drug delivery and elimination of cancer stem cells. These innovative therapies have fewer side effects compared to standard chemotherapy and address the stability issues associated with cancer immunotherapy. This review discusses various nanotheranostic systems and smart nanoparticles, their mechanisms of action, and their relevance in the current era.

Near-infrared-II photothermal ultra-small carbon dots promoting anticancer efficiency by enhancing tumor penetration

Summary: Improving the deep penetration of drugs in tumor sites is crucial for enhancing the therapeutic effect of tumors due to the unique tumor microenvironment. This study developed ultra-small permeable carbon dots (PCD) that can penetrate deep into tumor tissue without modification using near-infrared-II (NIR-II) window photothermal irradiation and exhibited good biocompatibility. The PCD showed multi-color fluorescence under visible light and photoacoustic signals under excitation, allowing for fluorescence and photoacoustic imaging for localization and distribution both in vitro and in vivo.

Recent trends of mesoporous silica-based nanoplatforms for nanodynamic therapies

Summary: Nanodynamic therapies using nanosensitizers triggered by various activators or reactions in tumor microenvironment have emerged as attractive strategies for treating deep-seated tumors. Mesoporous silica nanoparticles (MSNs) and MSNs-based hybrid nanoparticles are promising candidates for nanodynamic therapies due to their large surface area, high pore volume, biosafety, and easy surface modification. This review summarizes the antitumor mechanisms and latest progress in the construction, modification, and application of MSNs-based nanodynamic therapies, including photodynamic therapy, chemodynamic therapy, sonodynamic therapy, thermodynamic therapy, radiodynamic therapy, and electrodynamic therapy. The challenges and prospects for clinical applications of MSNs-based ROS-activated cancer treatments are also discussed.
