Lyotropic liquid crystals for parenteral drug delivery

Vivek P. Chavda et al.

Summary: The development of novel long-acting parenteral formulations based on lipid-based liquid crystals has been encouraged by the need for long-term treatments of chronic diseases. These liquid crystal systems, formed by amphiphilic molecules, spontaneously form liquid crystal nanoparticles in aqueous tissue environments, creating an in-situ long-acting sustained-release depot for extended treatment efficacy. This manuscript summarizes the different types of liquid crystals, recent formulation advancements, analytical evaluation, and therapeutic application of lyotropic liquid crystals in the field of parenteral sustained release drug delivery.

Dynamic light scattering analysis of size-selected graphene oxide 2D colloids fractioned via liquid crystal phase separation

Christina D. Abele et al.

Summary: Exfoliated platelets of graphene oxide (GO) can form nematic colloidal liquid crystal phases even at low concentrations. The precise analysis and control of GO sheet size distribution is crucial for scientific understanding and potential applications. This study presents a method to determine the mean effective width of GO platelets using depolarized dynamic light scattering and a model for circular disks. It also develops a simple fractionation protocol to prepare monodisperse fractions of GO sheets. These methods advance the fabrication of well-defined and size-selected GO-based systems.

An overview of PLGA in-situ forming implants based on solvent exchange technique: effect of formulation components and characterization

Tarek Metwally Ibrahim et al.

Summary: Due to low oral bioavailability of certain drugs, there is growing interest in parenteral administration to directly target drug absorption into the blood. Injectable in-situ forming implants (ISFIs) are gaining popularity compared to other sustained release systems, offering easier production, acceptable administration routes, reduced dosing frequency, and improved patient compliance. The use of biodegradable polymers like PLGA in ISFI systems allows for extended drug delivery through solvent exchange mechanisms, with drug release characterized by initial burst, diffusion processes, and polymeric degradation. Suitable solvent selection and consideration of PLGA hydrophobicity and molecular weights are crucial for optimizing drug release in ISFIs.
