Characterization of a glycoside hydrolase endolysin from Acinetobacter baumannii phage AbTZA1 with high antibacterial potency and novel structural features

Georgios E. Premetis et al.

Summary: This study identified a potential endolysin (AbLys1) with high selectivity towards Acinetobacter baumannii and characterized its enzymatic activity. The structure of AbLys1 revealed two helical domains and short sequences with predicted antimicrobial activity. Engineering studies demonstrated the importance of these sequences in the lytic activity of AbLys1 towards A. baumannii cells.

InterPro in 2022

Typhaine Paysan-Lafosse et al.

Summary: The InterPro database has been updated with new data content and website features, providing a more user-friendly access to protein sequence classification and functional domain identification. It has also integrated features from the retiring Pfam website and developed a card game to engage the non-scientific community. Furthermore, the database explores the benefits and challenges of using artificial intelligence for protein structure prediction.

Database resources of the national center for biotechnology information

Eric W. Sayers et al.

Summary: The National Center for Biotechnology Information (NCBI) produces a variety of online information resources for biology, including databases for nucleic acid sequences and life science journal citations. It provides search and retrieval operations for most of these data from 35 distinct databases, with E-utilities serving as the programming interface. Several resources received significant updates in the past year.

AI4AMP: an Antimicrobial Peptide Predictor Using Physicochemical Property-Based Encoding Method and Deep Learning

Tzu-Tang Lin et al.

Summary: Antimicrobial peptides (AMPs) have attracted drug developers as potential antibiotic substitutes, but discovering new candidates through wet-lab screening is inefficient and expensive. This study integrated up-to-date AMP data set and protein-encoding methods with a deep learning model to achieve high prediction accuracy, implemented in a user-friendly web server AI4AMP.

The Advantages and Challenges of Using Endolysins in a Clinical Setting

Ellen Murray et al.

Summary: Antibiotic-resistant pathogens are on the rise, posing a growing problem. Traditional antibiotics are no longer effective, creating a need for new approaches. Bacteriophage-derived proteins, such as endolysins, have shown promise in effectively targeting and destroying bacterial cells, particularly Gram-positive bacteria. This has led to a renewed interest in endolysin therapy and its potential as a new antimicrobial therapy, despite some regulatory and logistical challenges.

Mining of Gram-Negative Surface-Active Enzybiotic Candidates by Sequence-Based Calculation of Physicochemical Properties

Roberto Vazquez et al.

Summary: Phage (endo)lysins have emerged as a promising solution to the antibiotic resistance crisis, either as standalone therapeutics or in combination with traditional antibiotics. Advances in engineering approaches and understanding natural mechanisms have enabled the targeting of Gram-negative bacteria. Additionally, the identification of antimicrobial peptide-like regions within lysins has provided a potential source for novel antimicrobials.
