Adaptive exhaustion during prolonged intermittent hypoxia causes dysregulated skeletal muscle protein homeostasis

Summary: Nocturnal hypoxaemia, common in COPD patients, is associated with skeletal muscle loss and adverse clinical outcomes. This study investigated the adaptive cellular responses to prolonged intermittent hypoxia (PIH) and chronic hypoxia (CH). The results revealed shared perturbations in ribosomal and mitochondrial function, a sarcopenic phenotype, and mitochondrial oxidative dysfunction during both PIH and CH. These findings suggest an adaptive exhaustion and failure to restore homeostasis during normoxia. The physiological relevance was further confirmed in skeletal muscle of mice with COPD.

Cellular interplay in skeletal muscle regeneration and wasting: insights from animal models

Summary: Skeletal muscle wasting, whether related to ageing, disuse or chronic disease, is a crucial factor affecting quality of life and mortality. The cellular mechanisms responsible for increased catabolism in skeletal muscle cells are often unclear. Animal models, such as rodents, provide valuable insights into this dynamic process and its underlying mechanisms. Satellite cells, fibroblasts, immune cells, and other cell types play important roles in muscle regeneration and are altered in various muscle wasting conditions. Understanding these cellular alterations can lead to potential therapeutic strategies for muscle wasting diseases.

Additionally, COPD patients show upregulation of SR stress markers, disruption of myonuclear organization, and elevated levels of oxidative stress, all contributing to muscle weakness in advanced COPD.

Pathogenesis of sarcopenia and the relationship with fat mass: descriptive review

Summary: Age-associated obesity and muscle atrophy are closely linked and regulated by adipose tissue and skeletal muscle dysfunction. Adipose inflammation leads to fat redistribution and fatty infiltrations in muscles, causing decreased strength and functionality. Lipid accumulation induces mitochondrial dysfunction, increased production of reactive oxygen species, leading to lipotoxicity and insulin resistance. Muscle-secreted cytokines may exacerbate adipose tissue atrophy and chronic inflammation, establishing a vicious cycle of local hyperlipidaemia, insulin resistance, and inflammation, promoting the development of sarcopenic obesity.

Deaccelerated Myogenesis and Autophagy in Genetically Induced Pulmonary Emphysema

Summary: This study utilized a multiomics approach to analyze molecular changes during hyperammonemia in muscle tissues, revealing various types of responses including mitochondrial and oxidative dysfunction, disruption of protein homeostasis, and activation of oxidative stress pathways. Additionally, novel observations were made, such as an increase in expression of antiapoptotic proteins, increased calcium flux, and increased protein glycosylation during hyperammonemia. The data demonstrate complex molecular interactions during adaptive and maladaptive responses to cellular stress induced by hyperammonemia.
