Interaction of DWORF with SERCA and PLB as determined by EPR spectroscopy

Summary: Insufficient activity of SERCA, the integral transmembrane Ca2+ pump, contributes significantly to heart failure, a leading cause of death worldwide. The novel DWORF micropeptide has been identified as a muscle-specific SERCA effector that can reverse the inhibitory effects of PLB and activate SERCA independently. However, the structural basis for these functions has not yet been determined.

Ameloblastin promotes polarization of ameloblast cell lines in a 3-D cell culture system

Summary: Studies on animal models have shown that ameloblastin (AMBN) is involved in cell-matrix adhesion and cell polarization during enamel formation. In this study, an in vitro 3D cell culture model was developed to investigate the role of AMBN in cell polarization and morphology. The addition of AMBN to the culture matrix resulted in the clustering and elongation of ameloblast cell lines in a dose-dependent manner. AMBN plays a critical signaling role in the initiation of cell polarity by acting as a communicator between cell-cell and cell-matrix interactions.

The sequence encoded by exon 5 of ameloblastin affects amelogenin self-assembly, and their coassembly stabilizes amorphous calcium phosphate, enhancing our understanding of enamel matrix proteins' important role.

Coemergence of the Amphipathic Helix on Ameloblastin With Mammalian Prismatic Enamel

Summary: By analyzing Ambn sequences of 53 species, we found that a potential amphipathic helix (AH) within the sequence encoded by Exon 5 of Ambn appeared in species with prismatic enamel. Synthetic peptides derived from mammalian Ambn interacted with liposomes and caused liposome leakage, while peptides from species without prismatic enamel did not show similar effects. Our findings suggest that the AH motif is closely related to the emergence of enamel prismatic structure and is involved in Ambn's regulation of cell morphology and polarization genes.
