Therapeutic Targets and Emerging Treatments in Advanced Chondrosarcoma

Summary: Chondrosarcomas develop resistance to standard anticancer drugs, making it challenging to control the disease. New treatment approaches, such as molecule-targeting agents and immunotherapy, are needed. Recent studies have identified promising biomarkers and therapeutic targets for chondrosarcoma, and various molecule-targeting agents and immunotherapies have shown favorable antitumor activity in clinical studies. This review summarizes recent research on biomarkers and therapeutic targets, as well as clinical studies on the treatment of chondrosarcomas.

Genetics and epigenetics in conventional chondrosarcoma with focus on non-coding RNAs

Summary: This article reviews the important role of IDH 1/2 gene mutations in the molecular carcinogenesis of chondromas and chondrosarcomas. The mutations lead to the production of the non-physiological metabolite D-2HG, which inhibits a group of alpha-KG-dependent enzyme families, affecting the regulation of epigenetic factors. Additionally, ncRNAs are found to play a significant role in cancer development, as exemplified by the oncogenic functions of miRNAs in chondrosarcoma. Particularly in chondrosarcomas, the interactions between ncRNAs and alpha-KG-dependent epigenetic regulators contribute to the development of oncogenic traits.

Summary: BMS-986158, an experimental anticancer therapy, can block BET protein function to regulate cancer-related genes and inhibit tumor growth. This study evaluated the safety, tolerability, pharmacokinetics (PK), and pharmacodynamics of BMS-986158. Among the different dosing schedules tested, schedule A (5 days on, 2 days off) showed stable drug levels in the blood, preliminary anticancer effects, and a proportional PK profile.

Summary: Myelodysplastic Syndromes (MDS) affecting the elderly can progress to Acute Myeloid Leukemia (AML), with epigenetic alterations such as DNA methylation and chromatin modification playing a role in their initiation and progression. DNA hypomethylating agents like decitabine and azacitidine have shown efficacy in treating hematological malignancies, while the proteasome inhibitor bortezomib is used in multiple myeloma (MM) chemotherapy. Phase III clinical trials have demonstrated the potential of these drugs in prolonging survival and treating MDS and AML, although further research is needed to fully understand their mechanisms and effects.
