Cellular and Molecular Signatures of Oxidative Stress in Bronchial Epithelial Cell Models Injured by Cigarette Smoke Extract

Summary: Exposure of airway epithelium to environmental insults, such as cigarette smoke, leads to increased oxidative stress, which promotes the development of chronic lung diseases. Oxidative stress disrupts antioxidant defenses, affects cellular autophagy and survival regulatory mechanisms, leading to cell senescence. These events are exacerbated by inflammation driven by oxidative stress. Various models of bronchial epithelial cells are used to study the mechanisms and functions altered by cigarette smoke exposure and to test the efficacy of antioxidants. This review provides a comprehensive synthesis of in-vitro and ex-vivo studies from 2011 to 2021, investigating the molecular and cellular mechanisms triggered by cigarette smoke exposure in bronchial epithelial cells, commonly used experimental models, and the mechanisms of action of cellular antioxidant systems, as well as natural and synthetic antioxidant compounds.

NCOA4-Mediated Ferroptosis in Bronchial Epithelial Cells Promotes Macrophage M2 Polarization in COPD Emphysema

Summary: This study investigated the relationship between macrophage polarization and ferroptosis in COPD emphysema. The findings showed increased M2 macrophages and levels of matrix metalloproteinases (MMPs) in COPD patients, CS-exposed mice, and THP-M cells cocultured with CSE-treated HBE cells. Increased levels of NCOA4 and ferroptosis were also confirmed in COPD. The study suggests that NCOA4, by inducing an increase in M2 macrophages, plays a role in the pathogenesis of COPD emphysema through mediating ferroptosis.

Genetic tracing reveals transcription factor Foxp3-dependent and Foxp3-independent functionality of peripherally induced Treg cells

Summary: The role of Foxp3 in peripheral pTreg cells and the mechanisms supporting their differentiation remain poorly understood. This study used genetic tracing to identify microbiota-induced pTreg cells and found that many of their distinguishing features were Foxp3 independent. While Foxp3 was critical for the suppression of certain diseases, pTreg cells could suppress colonic effector T cell expansion in a Foxp3-independent manner. Thus, Foxp3 and the tolerogenic signals preceding and promoting its expression confer distinct facets of pTreg functionality.

Summary: The study investigated the expression and location of ACE2 in a cigarette smoke-exposed mouse model and human lung tissues, finding significant overexpression of ACE2 in bronchial and alveolar epithelial cells in smokers. Decreased ACE2 expression was observed in ex-smokers and correlated positively with HIF-1 alpha, iNOS, and 4-HNE levels.
