Nuclear factor Nrf2 promotes glycosidase OGG1 expression by activating the AKT pathway to enhance leukemia cell resistance to cytarabine

Summary: Chemotherapy resistance is a major challenge in AML treatment. Nrf2 plays a vital role in drug resistance of many tumors. The high expression of Nrf2 and OGG1 in AML patients is associated with disease progression. Upregulation of Nrf2 decreases sensitivity to cytarabine in leukemia cells, while downregulation of Nrf2 enhances chemosensitivity in drug-resistant cells.

DNMT3A R882H mutation promotes acute leukemic cell survival by regulating glycolysis through the NRF2/NQO1 axis

Summary: Studies have shown that acute myeloid leukemia (AML) cells with DNMT3A R882H mutation have increased proliferation capability. The mechanism behind this phenomenon remains unclear. This study found that the DNMT3A R882H mutation enhances glycolysis through activation of the NRF2/NQO1 pathway, which promotes AML cell proliferation. Inhibiting glycolysis can enhance the anticancer effects of daunorubicin in mutant cells.

Summary: mTOR activation is associated with resistance to GC-based chemotherapy in T-ALL. In this study, it was found that the mTOR inhibitor everolimus increased ROS levels, activated NRF2, induced mitochondrial inner membrane depolarization and apoptosis of T-ALL cells. The combination of everolimus and dexamethasone had a synergistic effect on killing T-ALL cells.

Summary: This study found that AML cells with DNMT3A R882H mutation exhibited high proliferative and anti-apoptotic activities, as well as resistance to daunorubicin treatment. The NRF2 pathway played an important role in the regulation of cell apoptosis, and the expression of NRF2 was regulated by DNMT3A R882H mutation through protein stability rather than methylation. Targeting NRF2 might be a potential therapeutic strategy for AML patients with DNMT3A R882H mutation.
