Age- and Sex-Dependent Changes of Free Circulating Blood Metabolite and Lipid Abundances, Correlations, and Ratios

Francesca Di Cesare et al.

Summary: This study investigated the variations in concentrations, correlations, and ratios of 202 free circulating blood metabolites and lipids with age. The results showed that certain metabolites and lipids were associated with age in a sex-dependent manner, with linoleic acid, alpha-linoleic acid, and carnitine being associated with age in women, and monoacylglycerols and lysophosphatidylcholines being associated with age in men. Furthermore, correlations among phosphatidylcholines tended to have a positive trend with age in women, while correlations among monoacylglycerols and lysophosphatidylcholines tended to have a negative trend with age in men. The ratios between molecular features also showed age-dependent changes, with the decanoyl-l-carnitine/lysophosphatidylcholine ratio decreasing with age in women, and the l-carnitine/phosphatidylcholine and l-acetylcarnitine/phosphatidylcholine ratios increasing with age in men.

Non-alcoholic fatty liver disease and increased risk of incident extrahepatic cancers: a meta-analysis of observational cohort studies

Alessandro Mantovani et al.

Summary: This meta-analysis found that NAFLD was significantly associated with an increased risk of developing gastrointestinal cancers (esophagus, stomach, pancreas, or colorectal cancers) by nearly 1.5-fold to 2-fold, as well as an increased risk of developing lung, breast, gynecological, or urinary system cancers by approximately 1.2-fold to 1.5-fold. These risks were independent of age, sex, smoking, obesity, diabetes, or other potential confounders.

Body mass index and adipose distribution have opposing genetic impacts on human blood traits

Christopher S. Thom et al.

Summary: Using Mendelian randomization, this study reveals opposite effects of genetically determined BMI and lipid levels on blood traits. Increased BMI affects multiple blood lineages, while increased truncal adipose distribution opposes BMI effects and increases blood cell formation and function.

Exercise training remodels subcutaneous adipose tissue in adults with obesity even without weight loss

Cheehoon Ahn et al.

Summary: Even without weight loss, 12 weeks of exercise training can alter adipose tissue structure and factors that regulate metabolism and the inflammatory signal pathway in adults with obesity.

Strength training alters the tissue fatty acids profile and slightly improves the thermogenic pathway in the adipose tissue of obese mice

Diego Gomes de Melo et al.

Summary: Short-term strength training was shown to decrease fatty acid levels and lipogenesis enzyme expression in subcutaneous adipose tissue (sWAT) of obese mice. Additionally, it increased the expression of browning-related genes. These effects were observed independently of changes in body weight.

Effects of exercise on NAFLD using non-targeted metabolomics in adipose tissue, plasma, urine, and stool

Ambrin Farizah Babu et al.

Summary: This study investigated the metabolic changes associated with non-alcoholic fatty liver disease (NAFLD) in response to high-intensity interval training (HIIT) intervention. The results showed that HIIT significantly improved clinical parameters of NAFLD patients, including fasting plasma glucose concentration and waist circumference. Metabolomics analysis revealed sample-type-specific changes in metabolite levels, with amino acids and their derivatives accumulating in adipose tissue and plasma, while decreasing in urine and stool. The study highlights the role of adipose tissue in responding to HIIT and suggests that altered amino acid metabolism in adipose tissue may play a critical role in improving fasting plasma glucose concentration.

Effects of exercise intensity on white adipose tissue browning and its regulatory signals in mice

Riku Tanimura et al.

Summary: The effects of high- or low-intensity exercises on the browning of white adipose tissue (WAT) were examined in this study. It was found that high-intensity exercise increased mitochondrial contents and heat production capacity in WAT, and also increased the protein expression level of fibroblast growth factor 21 (FGF21) in skeletal muscle.

Mortality in biopsy-confirmed nonalcoholic fatty liver disease: results from a nationwide cohort

Tracey G. Simon et al.

Summary: Population-based data in Sweden showed that all histological stages of NAFLD were linked with significantly increased overall mortality, and the risk escalated progressively with worsening histology. Excess mortality was mainly attributed to extrahepatic cancer and cirrhosis, while cardiovascular disease and HCC made modest contributions.

Summary: This study identified major risk factors for the development of hepatocellular carcinoma (HCC) in non-alcoholic fatty liver disease (NAFLD) patients. In non-cirrhotic NAFLD patients, older male patients with a history of smoking, diabetes, and elevated alanine aminotransferase had the highest risk of developing HCC.

Non-alcoholic fatty liver disease

Elizabeth E. Powell et al.

Summary: Non-alcoholic fatty liver disease (NAFLD) is a common global health issue with significant associations with metabolic syndrome and type 2 diabetes, leading to serious complications like cirrhosis and hepatocellular carcinoma. While cardiovascular disease and extrahepatic malignancy are the main causes of death in NAFLD patients, advanced liver fibrosis is a key prognostic marker for liver-related outcomes and overall mortality.
