Frontline Sodium Channel-Blocking Antiseizure Medicine Use Promotes Future Onset of Drug-Resistant Chronic Seizures

Chronic seizures induce sex-specific cognitive deficits with loss of presenilin 2 function

Summary: Patients with early-onset Alzheimer's disease (EOAD) are at higher risk of seizures, especially those with presenilin 2 (PSEN2) variants. Seizures can worsen cognitive function in AD, and the relationship between seizures and amyloid beta accumulation has been studied extensively. However, the impact of other factors on seizure susceptibility in EOAD is still not well understood. This study aimed to investigate the effects of loss of normal PSEN2 function and chronic seizures on cognitive function in aged mice. The results showed that chronic seizures significantly worsened hippocampus-dependent cognitive deficits in female PSEN2 KO mice, while loss of PSEN2 function alone did not affect cognitive performance.

Reductions in Hydrogen Sulfide and Changes in Mitochondrial Quality Control Proteins Are Evident in the Early Phases of the Corneally Kindled Mouse Model of Epilepsy

Summary: Epilepsy is a complex neurological disorder characterized by seizures, mitochondrial dysfunction, and neurodegeneration. This study investigated the changes in hydrogen sulfide (H2S) levels during seizures and the development of epilepsy. The results showed that plasma H2S levels were reduced after seizures and during the early phase of epilepsy. Furthermore, the expression of mitochondrial membrane integrity-related proteins increased during epilepsy development. These findings suggest that H2S may play a role in the pathogenesis of epilepsy.
