Involvement of aquaporin 5 in Sjo?gren?s syndrome

Clara Chivasso et al.

Summary: Sjo center dot gren's syndrome (SS) is a chronic autoimmune disease characterized by lymphoplasmacytic infiltration of salivary and lacrimal glands, leading to a decrease in tear and saliva production. Recent research has identified aquaporin-5 (AQP5) as a key player in saliva secretion and dysregulation of salivary gland epithelial cells in SS. Altered expression and localization of AQP5, as well as its protein partners, are considered new factors in the development of SS.

Cytosolic escape of mitochondrial DNA triggers cGAS-STING-NLRP3 axis-dependent nucleus pulposus cell pyroptosis

Weifeng Zhang et al.

Summary: Investigating the mechanisms of intervertebral disc degeneration (IVD) has revealed a potential treatment for limiting the progression of low back pain. IVD involves widespread inflammation and cell death within the discs, but the exact progression of the condition is unclear. Researchers in China have found that oxidative stress triggers the leakage of mitochondrial DNA (mtDNA) into the cellular fluid, which activates an immune response pathway and leads to the release of inflammatory cytokines. The activation of this pathway is directly linked to the severity of IVD, allowing for potential monitoring of the condition. Administering a drug to block mtDNA leakage in rats has been shown to slow the progression of IVD.

The potential role of resveratrol as supportive antiviral in treating conditions such as COVID-19-A formulator's perspective

Roy van Brummelen et al.

Summary: With the increased transmissibility and milder disease form of the omicron variant of COVID-19, there is a need to refocus on current treatment regimens. This includes considering safe, affordable, and available adjuvant treatments, as well as repurposing known drugs and substances. Resveratrol, a natural antioxidant, has shown potential as an antiviral and immune stimulant, reducing lung injury and pro-inflammatory cytokine release. However, new initiatives and collaborations are needed to fully unleash the potential of resveratrol in the pharmaceutical market.

Mitochondrial double-stranded RNAs as a pivotal mediator in the pathogenesis of Sj?gren's syndrome

Jimin Yoon et al.

Summary: This study found elevated levels of mitochondrial double-stranded RNAs (mt-dsRNAs) in the saliva and tears of Sjogren's syndrome (SS) patients, suggesting their involvement in the pathogenesis of the disease. The study also demonstrated that mt-dsRNAs may act as propagators of a pseudo-viral signal in the target tissue of SS.

Mitochondrial double-stranded RNAs govern the stress response in chondrocytes to promote osteoarthritis development

Sujin Kim et al.

Summary: This study reveals that mitochondrial double-stranded RNA (mt-dsRNA) plays a role in activating innate immune responses and contributing to the development of osteoarthritis (OA). The release of mt-dsRNA into the extracellular space and its activation of Toll-like receptor 3 at the plasma membrane are shown to be involved in this process. The findings suggest that targeting mt-dsRNA could be a potential strategy for chondroprotective intervention.

Summary: Sjogren's syndrome is a common autoimmune disease characterized by dry mouth and eyes, and can involve multiple systems. Previous research has focused on the role of B cells and T cells in the pathogenesis and progression of Sjogren's syndrome. However, recent studies have identified monocytes as a third player connecting innate and adaptive immunity in this disease.

Mitochondria-Induced Immune Response as a Trigger for Neurodegeneration: A Pathogen from Within

Marta Luna-Sanchez et al.

Summary: The symbiosis between mitochondria and the ancestor of the eukaryotic cell is crucial for cellular complexity and survival, but compromised mitochondrial integrity can lead to unwanted cellular immune responses. Mitochondrial nucleic acids like mtDNA and mtRNA can interact with cytoplasmic sensors, triggering inflammation. Mitochondria-driven inflammation may serve as a potential therapeutic target for neurodegenerative and primary mitochondrial diseases.
