Infiltrating CD8+T cells and M2 macrophages are retained in tumor matrix tracks enriched in low tension fibronectin fibers

Charlotte M. Fonta et al.

Summary: Immunostainings of mouse breast tumors reveal that tumor matrix tracks, with immunosuppressive functions, are surrounded by an endothelial-like basement membrane and contain collagen and fibronectin fibers. These tracks form in early-stage tumors and mature over time, and their maturation depends on host-derived tenascin-C (TNC). Tumor infiltrating leukocytes, mainly M2 macrophages and CD8+ T cells, are retained in early-stage tumor tracks but reduce in number after track maturation.

Modulating tenascin-C functions by targeting the MAtrix REgulating MOtif, MAREMO

Thomas Loustau et al.

Summary: The extracellular matrix molecule Tenascin-C (TNC) promotes cancer and chronic inflammation through various mechanisms and contributes to an immune suppressive tumor microenvironment. Research has identified a highly conserved MAtrix REgulating MOtif (MAREMO) or M-motif in fibronectin and TNC. Mimetic peptides of the M-motif have been found to inhibit the functions of TNC.

Tenascin-C immobilizes infiltrating T lymphocytes through CXCL12 promoting breast cancer progression

Devadarssen Murdamoothoo et al.

Summary: The study found that tenascin-C inhibits CD8 T cell infiltration by inducing CXCL12, which may contribute to the low response rates of immune checkpoint therapy in cancer treatment. Blocking the CXCR4 receptor of CXCL12 enhances macrophage and CD8 T cell infiltration, reducing tumor growth and metastasis.

Summary: Despite major breakthroughs in cancer immunotherapy, only a small percentage of patients benefit significantly. Chronic inflammation in the tumor microenvironment plays a key role in tumor immunosuppression, with tumor-associated ECM identified as a major obstacle to successful cancer immunotherapy cases.

Expression of CCL2/CCR2 signaling proteins in breast carcinoma cells is associated with invasive progression

Wei Bin Fang et al.

Summary: DCIS is the most common type of pre-invasive breast cancer in women, with most cases unlikely to progress to invasive breast cancer. Increased CCL2/CCR2 signaling in breast tissue is associated with DCIS progression, potentially serving as a signature to predict the likelihood of DCIS progressing to IDC.
