Central opioid receptors mediate morphine-induced itch and chronic itch via disinhibition

Zilong Wang et al.

Summary: The study demonstrates that intrathecal opioids can induce itching by acting on MOR on spinal inhibitory interneurons, resulting in disinhibition of the spinal itch circuit. In addition, spinal ablation of GRPR(+) neurons can reduce opioid-induced itch, while intrathecal NPY administration can suppress itch response.

CXCL10/CXCR3 Signaling in the DRG Exacerbates Neuropathic Pain in Mice

Yan-Fang Kong et al.

Summary: Chemokines and their receptors play a role in chronic pain pathogenesis, with CXCR3 and CXCL10 specifically influencing neuronal excitability in the dorsal root ganglion. Activation of CXCR3 by CXCL10 leads to p38 and ERK activation in DRG neurons, enhancing neuronal excitability and contributing to neuropathic pain maintenance.

The Phosphorylated Form of the Histone H2AX (γH2AX) in the Brain from Embryonic Life to Old Age

Adalberto Merighi et al.

Summary: The gamma phosphorylated form of histone H2AX, known as gamma H2AX, was discovered over 40 years ago and has been found to play a crucial role in the cellular response to DNA damage. It is involved in various cellular functions in both normal and pathological cells. Particularly, its involvement in the central nervous system, from immature to mature neurons, highlights its importance in neurogenesis and neuronal cell death, showcasing its pleiotropic nature with diverse functions from embryonic life to old age.

Potassium channels and autism spectrum disorder: An overview

Peipei Cheng et al.

Summary: Autism spectrum disorder (ASD) is a group of neurodevelopmental disorders characterized by impaired social interaction and communication, and restricted, repetitive behaviors. Potassium channels play a key role in regulating neuronal excitability, and ASD susceptibility genes associated with potassium channels may contribute to altered brain function in ASD pathogenesis.

Peripheral Voltage-Gated Cation Channels in Neuropathic Pain and Their Potential as Therapeutic Targets

Sascha R. A. Alles et al.

Summary: The persistence of increased excitability and spontaneous activity in injured peripheral neurons is crucial for neuropathic pain development and many forms of neuropathic pain. However, the therapeutic potential of peripherally acting channel modulators is promising but their clinical performance has been disappointing.

Epigenetic modifications in neuropathic pain

Danzhi Luo et al.

Summary: Neuropathic pain is a common symptom in diseases of the somatosensory nervous system and is severely impacted by epigenetic modifications. These modifications, including DNA methylation, histone modification, non-coding RNA, and RNA modification, play a crucial role in regulating gene expression, function, and crosstalk between genes and the environment, offering novel targets for NP therapy.

Stepwise crosstalk between aberrant Nf1, Tp53 and Rb signalling pathways induces gliomagenesis in zebrafish

Juanjuan Luo et al.

Summary: The molecular pathogenesis of glioblastoma is critical for human gliomagenesis, with RTK/Ras/PI3K, RB, and TP53 pathways playing key roles. Using transgenic zebrafish with single or multiple gene deletions, researchers induced gliomagenesis and found that nf1 mutation alone had low efficiency, but combined deletions of nf1;tp53 and rb1;tp53 resulted in higher tumor incidences with advanced histology. Bioinformatics analysis revealed alterations in critical pathways like RTK/Ras/PI3K, cell cycle, and focal adhesion, highlighting their importance in gliomagenesis initiation and development. Tumors in zebrafish models showed significant similarities to human gliomas, and temozolomide treatment effectively suppressed gliomagenesis, mimicking responses seen in clinical glioma treatment. This study provides a potential tool for genetically studying gliomagenesis and screening targeted anti-tumor compounds for glioma treatment.
