Antioxidant and Anti-Inflammatory Effect of Cinnamon (Cinnamomum verum J. Presl) Bark Extract after In Vitro Digestion Simulation

Stefania Pagliari et al.

Summary: Cinnamon bark is widely used for its organoleptic features in food and has been found to have beneficial effects on human health. Cinnamon extracts contain antioxidant, anti-inflammatory, and anticancer biomolecules. This study evaluated the antioxidant and anti-inflammatory bioactivity of digested cinnamon extract and found that the digestive process reduces the polyphenol content while maintaining other bioactive compounds and functional effects at the cellular level.

New and Emerging Treatments for Inflammatory Bowel Disease

Masaaki Higashiyama et al.

Summary: This paper provides an overview of the current treatment status and future prospects for inflammatory bowel disease (IBD). While there are already many new treatment options available, there is still a need for better efficacy and fewer side effects in some refractory cases. Promising treatments with new perspectives are expected to emerge in the future.

AP2M1 mediates autophagy-induced CLDN2 (claudin 2) degradation through endocytosis and interaction with LC3 and reduces intestinal epithelial tight junction permeability

Ashwinkumar Subramenium Ganapathy et al.

Summary: This study revealed that increased clathrin-mediated endocytosis under starvation conditions promotes degradation of CLDN2 and enhances the TJ barrier. The association of CLDN2 with clathrin and AP2 as well as LC3 and lysosomes increased upon starvation, indicating the role of clathrin-mediated endocytosis in autophagy-induced CLDN2 degradation.

The Increase of miR-195-5p Reduces Intestinal Permeability in Ulcerative Colitis, Modulating Tight Junctions' Expression

Viviana Scalavino et al.

Summary: Defects in intestinal epithelial barrier functions are characteristic of inflammatory conditions such as Inflammatory Bowel Disease (IBD). This study found that miR-195-5p can regulate the expression of CLDN2 and other tight junction proteins under inflammatory conditions. It plays an important role in the integrity of the intestinal barrier and epithelial restoration.

Secretory autophagy maintains proteostasis upon lysosome inhibition

Tina A. Solvik et al.

Summary: The endolysosome system is involved in autophagic degradation and secretory pathways, including the release of extracellular vesicles and particles (EVPs). This study reveals a secretory autophagy pathway that is upregulated during endolysosome inhibition and mediates the release of autophagic cargo receptors via EVPs. This secretion process is regulated by multiple ATGs and the small GTPase Rab27a.

Characterization of the Biological Activities of a New Polyphenol-Rich Extract from Cinnamon Bark on a Probiotic Consortium and Its Action after Enzymatic and Microbial Fermentation on Colorectal Cell Lines

Alessandra De Giani et al.

Summary: Cinnamon polyphenols have antimicrobial activity against Staphylococcus aureus and Bacillus subtilis, but this activity is lost after in vitro digestion. However, they exhibit strong prebiotic potential for probiotic strains. Additionally, the secondary metabolites produced from cinnamon extract or its digested form have protective effects against colorectal tumor cells.

Coumarin Derivatives in Inflammatory Bowel Disease

Luiz C. Di Stasi

Summary: Inflammatory bowel disease is a chronic inflammatory disease without definitive pharmacological treatment. The unclear etiology limits the development of new drugs, leading to high rates of refractory patients. Natural products, such as coumarins, show potential as alternative therapies in IBD.

The Pathways Underlying the Multiple Roles of p62 in Inflammation and Cancer

Paulina Hennig et al.

Summary: P62 is a highly conserved adaptor protein involved in important cellular processes, regulating tissue homeostasis, inflammation, and diseases like cancer. Through interactions with multiple partners, P62 influences key pathways and acts as a central signaling hub for pro- and anti-inflammatory pathways. Its roles in autophagy, inflammation, and cancer highlight its potential as a target for therapeutic strategies in disease progression.
