HSP90 inhibitors and cancer: Prospects for use in targeted therapies (Review)

Summary: Heat shock protein 90 (HSP90) is a crucial chaperone protein that regulates signaling pathways and corrects misfolded proteins in cancer cells. Inhibition of HSP90 chaperone machinery has shown potential for inhibiting tumor survival, proliferation, invasion, and migration. Although several HSP90 inhibitors have been developed, their use as clinical treatments is currently limited by drug resistance, dose-limiting toxicity, and poor pharmacokinetic profiles. However, novel HSP90-targeted compounds are continuously being discovered and tested for their anticancer efficacy, and combination therapies with chemotherapy, targeted agents, or immunotherapy have shown improved antitumor effects. This review discusses the effects of HSP90 inhibitors on managing cancer and summarizes previous and novel HSP90-based therapeutic strategies in cancer treatment, proposing prospective candidates for future evaluation as cancer treatments.

TELO2-related syndrome (You-Hoover-Fong syndrome): Description of 14 new affected individuals and review of the literature

Summary: You-Hoover-Fong syndrome (YHFS) is an autosomal recessive condition caused by pathogenic variants in the TELO2 gene. It is characterized by global developmental delay, intellectual disability, microcephaly, dysmorphic facial features, ocular involvement, movement disorder, hypertonia and spasticity, and abnormal sleep pattern. Other features include poor growth, cleft palate, cardiac malformations, epilepsy, scoliosis, and hearing loss. This study describes 14 new individuals with YHFS and provides additional support of the phenotype recognized previously. A review of the literature including follow-up data on four previously reported individuals with YHFS is also presented.

TTI1 promotes non-small-cell lung cancer progression by regulating the mTOR signaling pathway

Summary: The expression and potential value of TELO2-interacting protein 1 (TTI1) in non-small-cell lung cancer (NSCLC) patients were investigated in this study. The results showed that TTI1 was highly expressed in NSCLC tissues and correlated with tumor size, tumor stage, and lymphatic metastasis. In vitro experiments demonstrated that TTI1 upregulation promoted cell invasion, metastasis, viability, and proliferation in NSCLC cells. Mechanistically, TTI1 was found to regulate mTOR activity, which plays a crucial role in tumor progression. Furthermore, the combination of TTI1 and Ki67 was identified as a valuable molecular biomarker for the prognosis and recurrence of NSCLC patients.

Structure of the human ATM kinase and mechanism of Nbs1 binding

Discovery of small-molecule inhibitors of RUVBL1/2 ATPase

Summary: RUVBL1 and RUVBL2 are highly conserved AAA ATPases that play a significant role in cancer progression. This study utilized docking-based virtual screening to identify compounds that inhibit the RUVBL1/2 complex. Seven compounds were found to have inhibitory activity in enzymatic and cellular assays. A series of pyrazolo[1,5-a]pyrimidine-3-carboxamide analogs were synthesized based on compound 15, which showed good potential for structural manipulation. Analysis of the structure-activity relationship revealed the importance of the benzyl group on R2 and the aromatic ring-substituted piperazinyl on R4 for inhibitory activity. Compound 18 exhibited the strongest inhibition and showed potential anticancer activity in multiple cell lines. Proteomic analysis identified cellular proteins dysregulated by compounds 16, 18, and 19. These findings suggest that compound 18 could serve as a starting point for structural modifications to improve potential therapeutic molecules.

Assembly mechanism of early Hsp90-Cdc37-kinase complexes

Summary: This study investigates the recruitment and loading mechanism of protein kinases to the Hsp90-Cdc37 complex, which is the first step in Hsp90-mediated chaperoning. The results show that the conformational dynamics of all partners are critical for the loading mechanism, and that Cdc37 plays a role in sensing clients by stabilizing the preexisting partially unfolded client state. These findings reveal the regulatory mechanism of molecular chaperones in maintaining protein homeostasis.

This highlights the importance of dedicated chaperones and interactions between subunits in the biogenesis of large complexes.

Summary: Smith-Kingsmore Syndrome is a rare neurodevelopmental disorder caused by mutations in the MTOR gene, leading to excessive growth of the head and brain, developmental delays, and intellectual disabilities. Research has identified novel MTOR variants as potential pathogenic factors in SKS, explored their impact on cellular function, and highlighted that mutations deep within the 3D structure of the MTOR protein are more likely to cause disease. Incorporating 3D properties of MTOR gene changes may aid in predicting pathogenicity.
