Summary: A new paradigm in nanomedicine has emerged, shifting from cell therapy to acellular extracellular vesicle (EV) therapy. However, technical limitations remain in the large-scale production of clinical-grade EV. In this study, a method of preparing vesicle-free hPL (EV-free hPL) and FCS (EV-free FCS) was developed using tangential flow filtration (TFF), providing a new option for producing large amounts of EV from any mammalian cells.

Summary: Extracellular vesicles (EVs) secreted by nucleated cells, especially endothelial progenitor cell-derived EVs (EPC-EVs), have shown beneficial effects in various diseases such as cardiovascular and lung disorders, accumulating at injury sites with angiogenic and restorative effects surpassing their ancestors. This review article focuses on the biogenesis, delivery routes, and long-term storage methods of EPC-EVs, highlighting their anti-inflammatory, angiogenic, and tissue protection effects in diseases, and discussing the therapeutic potential of EPC-EVs in clinical applications.

Summary: Mesenchymal stem cells and endothelial progenitor cells have unique immunoregulatory features, including proangiogenic functions and the ability to suppress the immune response, specifically T cell proliferation, activation, and cytokine production. However, the effectiveness of their immunosuppressive function can be influenced by various factors such as extraction sources, age/passage, expression of different markers, secretion of bioactive mediators, etc. Additionally, a comparison between the immunological interaction of mesenchymal stem cells and endothelial progenitor cells with T cells has not yet been published.
