Replication stress triggered by nucleotide pool imbalance drives DNA damage and cGAS-STING pathway activation in NAFLD

Romain Donne et al.

Summary: This study investigated the mechanisms of DNA damage response during NAFLD and found that nucleotide pool imbalance is the key driver of replication stress (RS), and DNA damage also activates the cGAS/STING pathway, promoting disease progression.

GABA regulates IL-1b production in macrophages

Jian Fu et al.

Summary: The study demonstrates that GABA plays a role in regulating macrophage maturation and inflammation. It enhances certain signaling pathways and reduces the formation of inflammatory complexes, leading to a decrease in pro-inflammatory responses. GABA also shows potential in alleviating sepsis and obesity in mice.

Effects of GABAB receptor positive allosteric modulator BHF177 and IRS-1 on apoptosis of hippocampal neurons in rats with refractory epilepsy via the PI3K/Akt pathway

Peng Wang et al.

Summary: The effects of the GABA(B) receptor positive allosteric modulator BHF177 on refractory epilepsy (RE) were investigated in this study. The results showed that BHF177 reduced seizure intensity and frequency, and decreased apoptosis and inflammatory response. Furthermore, BHF177 activated GABA(B) receptors and suppressed apoptosis in hippocampal tissues through the IRS-1/PI3K/Akt axis. These findings suggest a new therapeutic approach for RE.

Combined use of GABA and sitagliptin promotes human β-cell proliferation and reduces apoptosis

Wenjuan Liu et al.

Summary: The combination of GABA and Sita in human beta-cells showed superior therapeutic benefits in terms of increasing beta-cell counts, improving blood glucose levels, and enhancing beta-cell proliferation while reducing apoptosis. This synergistic effect is likely due to an enhancement of beta-cell proliferation and a decrease in apoptosis.

Common molecular mechanisms of SLC6A1 variant-mediated neurodevelopmental disorders in astrocytes and neurons

Felicia Mermer et al.

Summary: SLC6A1 gene variants are associated with a spectrum of epilepsy syndromes and neurodevelopmental disorders. The major disease mechanism appears to be impaired transporter protein trafficking and surface expression, leading to reduced GABA uptake. Abnormal subcellular localization, endoplasmic reticulum retention, and decreased cell surface expression of variant GAT-1 proteins are common findings, suggesting that boosting transporter function could be beneficial for therapeutic development.
