Extracellular Non-Coding RNAs in Cardiovascular Diseases

Zeyidan Jiapaer et al.

Summary: Cardiovascular diseases (CVDs) remain the leading cause of death worldwide despite advances in healthcare. Extracellular vesicles (EVs), particularly the RNA cargo, are recognized as promising targets for diagnosis and therapy in CVDs. Various types of non-coding RNAs (ncRNAs), such as small ncRNAs, circular RNAs, and long ncRNAs, have been found to modulate biological processes in recipient cells. This review summarizes recent research on the origin, carriers, functional mechanisms, and pathological roles of extracellular ncRNAs in different CVDs, and discusses the challenges and developments in using them as biomarkers or therapeutic targets.

ADSC-derived exosomes attenuate myocardial infarction injury by promoting miR-205-mediated cardiac angiogenesis

Tingting Wang et al.

Summary: This study investigated the protective effects of exosomes from adipose tissue-derived mesenchymal stem cells (ADSC-Exos) on myocardial infarction (MI)-treated mice. ADSC-Exos were found to alleviate cardiac injury, promote cardiac function recovery, and inhibit cardiomyocyte apoptosis through the miRNA-205 signaling pathway. These findings suggest that ADSC-Exos containing miRNA-205 have therapeutic potential for MI-induced cardiac injury.

Role of MSC-derived small extracellular vesicles in tissue repair and regeneration

Bruna Andrade Aguiar Koga et al.

Summary: Mesenchymal stem cells (MSCs) secrete vesicles known as extracellular vesicles (EVs), which play a crucial role in tissue homeostasis, repair, and therapeutic applications. MSC-derived EVs have advantages such as low immunogenicity and the ability to pass through barriers, making them potential drug delivery agents for various diseases. Extensive in vitro studies have been conducted to understand the mechanisms and signaling pathways of EVs. Clinical trials are underway to explore the therapeutic potential of EVs in Alzheimer's disease, retina degeneration, and COVID-19. This review provides an overview of different EV isolation methods and highlights their therapeutic applications in tissue repair and regeneration.

Xue-ling Su et al.

Summary: In this study, the effects of caspase-1 inhibitor VX765 on the expression of Cx43 and cell-to-cell communication after myocardial infarction (MI) were investigated. The results showed that VX765 treatment significantly decreased the infarct area, alleviated cardiac dysfunction and remodeling, and raised Cx43 levels in the heart after MI. In addition, VX765 treatment reversed the downregulation of Cx43 expression and improved intercellular communication in vitro.

De novo DNA methylation induced by circulating extracellular vesicles from acute coronary syndrome patients

Concetta Schiano et al.

Summary: DNA methylation is associated with cardiovascular diseases (CVDs). This study found that circulating extracellular vesicles (EVs) from acute coronary syndrome (ACS) patients showed increased levels of DNA methyltransferases (DNMTs) and reduced Ten-eleven translocation (TET) genes. By treating lymphocytes from healthy individuals with EVs, it was discovered that DNMTs carried by EVs play an important role in DNA methylation. Furthermore, ACS-EVs were found to regulate signaling pathways and gene expression in target cells.

Tumor endothelial cell-derived extracellular vesicles contribute to tumor microenvironment remodeling

Jian Gao et al.

Summary: Cancer progression involves key steps such as angiogenesis, and extracellular vesicles (EVs) released by tumor cells and other cells in the tumor microenvironment (TME) play a role in modulating and maintaining favorable conditions for tumor growth. EVs derived from endothelial cells (ECs) have important roles in tumor angiogenesis and can also activate tumor cells, facilitating extracellular matrix (ECM) remodeling and providing additional nutrients to tumor cells. Therefore, targeting EV communication between cancer cells and ECs could be an effective therapeutic approach for controlling cancer progression.

Extracellular vesicles as central regulators of blood vessel function in cancer

Fernanda G. Kugeratski et al.

Summary: Blood vessels play a crucial role in tumor growth and cancer cell dissemination. However, abnormalities in tumor vasculature can contribute to an aggressive tumor microenvironment and reduce the effectiveness of cancer therapies. Recent studies have shown that extracellular vesicles (EVs) derived from cancer cells can alter the function of endothelial cells in blood vessels, leading to sustained angiogenic signaling in tumors. Targeting these EV-mediated mechanisms may provide new strategies to normalize tumor vasculature.
