The researchers identified serum response factor (SRF) as a mediator of OPC proliferation in response to young CSF, and fibroblast growth factor 17 (Fgf17) as a key target for restoring oligodendrocyte function in the ageing brain.

Human Forebrain Organoid-Derived Extracellular Vesicle Labeling with Iron Oxides for In Vitro Magnetic Resonance Imaging

Summary: Extracellular vesicles (EVs) play significant roles as intracellular mediators, disease biomarkers, and therapeutic agents. Superparamagnetic iron oxide (SPIO) nanoparticles have been shown to improve the targeting ability of EVs. In this study, ultrasound sonication was used to load ultrasmall SPIO (USPIO) nanoparticles onto EVs derived from induced forebrain neural progenitor cortical organoids (iNPCo). The labeled EVs exhibited higher MRI contrast in agarose gel phantoms.

Phenotypic, metabolic, and biogenesis properties of human stem cell-derived cerebellar spheroids

Engineering extracellular vesicles by three-dimensional dynamic culture of human mesenchymal stem cells

Summary: This study presents a promising strategy for scalable production of high-quality extracellular vesicles (EVs) from human mesenchymal stem cells (hMSCs) using 3D aggregates grown under wave motion. The 3D hMSC aggregates promote activation of different pathways involved in EV secretion. Comparison with 2D hMSC-EVs reveals that the quantity of 3D hMSC-EVs is significantly enhanced, with smaller sizes, higher expression of miR-21 and miR-22, and an altered protein cargo. Functionally, 3D hMSC-EVs rejuvenate senescent stem cells and exhibit enhanced immunomodulatory potentials.

Cerebellar Differentiation from Human Stem Cells Through Retinoid, Wnt, and Sonic Hedgehog Pathways

Summary: This study investigated the synergistic effects of retinoic acid, Wnt activator, and sonic hedgehog activator on cerebellar patterning of human-induced pluripotent stem cell (hiPSC) spheroids and organoids. The cells also exhibit electrophysiological characteristics using whole-cell patch clamp recording, especially demonstrating Purkinje cell electrophysiology.

These EVs from CP induced pro-inflammatory effects and inhibiting EV production with GW4869 protected against cognitive decline induced by A beta O.
