Label-Free Characterization of Macrophage Polarization Using Raman Spectroscopy

Max Naumann et al.

Summary: Macrophages, important cells of the immune system, can be classified into different subtypes based on their function and phenotype. In this study, Raman spectroscopic imaging was used to visualize and discriminate between different human macrophage phenotypes in a non-destructive and label-free manner, which is important for studying their contribution to various pathologies.

Skeletal infections: microbial pathogenesis, immunity and clinical management

Elysia A. Masters et al.

Summary: This review explores the microbial pathogenesis, immunity, and clinical management of bone infections, highlighting osteomyelitis as a significant risk in orthopaedic surgery. It discusses the mechanisms of persistence and immune evasion in Staphylococcus aureus infection of the skeletal system, as well as features of other osteomyelitis-causing pathogens. The article also describes the clinical management of skeletal infection, including osteomyelitis classification and strategies for treating skeletal infections with emerging technologies.

A Journey into Animal Models of Human Osteomyelitis: A Review

Gabriele Meroni et al.

Summary: Osteomyelitis is a bone infection that can spread through hematogenous, contiguous, or direct inoculation routes. Animal models can be used to study osteomyelitis, and different species are chosen for research. The clinical manifestation of hematogenous osteomyelitis varies due to multiple factors, but these variables can be controlled and tested through animal models.

Raman microspectroscopy reveals unsaturation heterogeneity at the lipid droplet level and validates an in vitro model of bone marrow adipocyte subtypes

Josefine Tratwal et al.

Summary: BMAds are the most abundant stromal component of adult human bone marrow, consisting of two types: rBMAds and cBMAds, with distinct characteristics. An in vitro model of BM adipogenesis with differential rBMAd and cBMAd-like characteristics has been developed, showing differences in fatty acid composition and desaturase expression. This model is useful for discriminating cBMAds from rBMAd and studying adipocyte differentiation.

Raman microscopic spectroscopy as a diagnostic tool to detect Staphylococcus epidermidis in bone grafts

A. Wurm et al.

Summary: Raman microscopic spectroscopy is a promising tool for detecting Staphylococcus epidermidis in human bone grafts, with distinct diagnostic spectra being able to differentiate between infected and non-infected bone samples. Further studies in human tissues are needed to explore its full potential.

Emerging electron microscopy and 3D methodologies to interrogate Staphylococcus aureus osteomyelitis in murine models

Karen L. de Mesy Bentley et al.

Summary: Recent breakthroughs in understanding orthopaedic infections have been made through advances in transmission electron microscopy imaging of murine models, particularly in revealing the colonization process of Staphylococcus aureus in the bone networks. Three-dimensional reconstruction highlighted adjacent uninfected canaliculi, suggesting viable osteocytes may respond and resist infection in chronic osteomyelitis, opening new research directions.

Brillouin and Raman Micro-Spectroscopy: A Tool for Micro-Mechanical and Structural Characterization of Cortical and Trabecular Bone Tissues

Martina Alunni Cardinali et al.

Summary: This study explores the potential of using Brillouin and Raman micro-spectroscopic (BRmS) technique for studying bone tissue, demonstrating its capability in discovering morpho-mechanics of bone cells, extracellular matrix, and marrow constituents. The results reveal specific alterations in morphology and tissue chemical composition in different anatomical regions, which significantly affect bone mechanical properties and provide a potentially extendable method for other important biomedical issues.

Correlation of crystal violet biofilm test results of Staphylococcus aureus clinical isolates with Raman spectroscopic read-out

Christina Ebert et al.

Summary: The study correlated fast Raman spectroscopic read-out of clinical Staphylococcus aureus isolates with their biofilm-forming characteristics, revealing significant spectral differences between high and low biofilm performers in the fingerprint region between 750 and 1150 cm(-1). Partial least square regression (PLSR) analysis showed a slight linear correlation of the Raman spectra with biofilm OD values, highlighting spectral differences in Raman bands representing nucleic acids, carbohydrates, and proteins. This suggests a potential application of Raman spectroscopy as a fast prediction tool for biofilm formation of bacterial strains directly after isolation, aiding clinicians in making timely and adapted therapeutic decisions.

Staphylococcus aureus Cell Wall Biosynthesis Modulates Bone Invasion and Osteomyelitis Pathogenesis

Elysia A. Masters et al.

Summary: Invasion of the osteocyte lacuno-canalicular network by Staphylococcus aureus is a novel mechanism in chronic osteomyelitis, where cell wall synthesis machinery and surface adhesins play critical roles. Specifically, cell wall synthesis proteins are key modulators of osteomyelitis pathogenesis, while surface adhesins have minimal effects. Transmission electron microscopy imaging showed that pbp3 deletion was associated with decreased invasion of canaliculi in vivo.

Innovations in osteomyelitis research: A review of animal models

Kylie M. Roux et al.

Summary: Osteomyelitis is a growing concern in modern healthcare due to antibiotic resistance, and current treatments are limited in their effectiveness and can lead to recurrence. Developing safer and more effective treatment options is necessary, and animal models are essential for testing treatment regimens. However, establishing a model that accurately reflects the clinical disease remains a challenge for scientists.

Summary: The study revealed that in mice, fracture-related infection (FRI) leads to osteolysis, the formation of staphylococcal abscess communities (SACs), and an immunosuppressive environment. Mice inoculated with S. aureus showed abnormal bone healing and severe osteolysis.
