Summary: Recent advances in molecular subtyping of gastric cancer have identified new clinically relevant biomarkers and potential therapeutic targets. Checkpoint inhibitors have also expanded treatment options, showing promising results in certain subgroups of gastric cancer patients. However, cytotoxic chemotherapy remains the only choice for patients who do not have these biomarkers. This study aims to investigate the prevalence, clinicopathologic features, and treatment outcomes of molecular subtypes in a new-diagnosed cohort of Western gastric cancer patients.

Summary: Despite efforts made, glioblastoma remains a deadly primary brain cancer with no cure. The peritumoural brain zone (PBZ), the apparently normal area surrounding the tumour, has shown potential in the tumour's maintenance and progression. This study aims to better understand PBZ and identify genes that may play a role in its malignant transformation.

Summary: Despite recent advances in treatment approaches, cancer remains a major cause of death worldwide. The restoration of tumor immune surveillance has shown promising results in overcoming resistance to conventional therapies and improving the clinical management of advanced disease. However, only a fraction of patients benefits from cancer immunotherapy, due to mechanisms that allow tumors to escape immune surveillance. Research on drug repositioning has suggested that anthelmintic drugs, in addition to their antineoplastic effects, have significant immunomodulatory actions on specific subsets of immune cells and pathways. This review examines the impact of anthelmintic drugs on host immunity and their potential implications in cancer immunotherapy.

Summary: The aberrant expression of PD-L1 and mechanisms of immune evasion in cancer cells are reviewed. The potential of immunotherapy in treating head and neck squamous cell carcinoma and strategies for improving treatment outcomes, such as identifying treatment response biomarkers and exploring new pharmacological combinations and molecules, are summarized.

Summary: This study fills the information gap regarding the role of ICIs in NSCLC patients with other driver mutations and provides a molecular landscape of clinically relevant oncogenic drivers in PD-L1 positive NSCLC patients. The results demonstrate that some patients with a PD-L1 tumor proportion score of 50% or higher and concomitant gene alterations may benefit from ICIs treatment, even in the presence of a KRAS gene alteration.

Summary: This study evaluated the usefulness of liquid biopsy in the clinical management of advanced-stage oncogene-addicted non-small-cell lung adenocarcinomas. The results showed that liquid biopsy could detect disease progression earlier and identify new mutations responsible for treatment resistance. The study provided algorithms for the use of liquid biopsy in diagnosis and therapy monitoring.

Summary: This review article discusses the clinical features and common treatment approaches for hepatocellular carcinoma (HCC). Targeted nanoparticle-based therapies for HCC, which aim to improve drug delivery and overcome drug resistance, show promise by targeting glypican 3 (GPC3) as a tumor-associated antigen. The use of nanoparticles conjugated with anti-GPC3 agents may also reduce severe side effects caused by chemotherapy.

Summary: Radiotherapy is an effective and targeted treatment option for many cancer types, but radioresistance remains a clinical challenge. Recent studies suggest that mechanisms related to mitochondrial changes and metabolic remodeling play an important role in the development of radioresistance. This article discusses the main contributors to acquired cellular radioresistance and their relation to mitochondrial changes, and summarizes some recently proposed mitochondria-targeting strategies to overcome radioresistant phenotypes in cancer.

Summary: Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic cancer, with low survival rates and a majority of patients already in advanced stages. Current treatment options for PDAC are limited, highlighting the need for targeted therapies that can differentiate between cancerous and non-cancerous cells. Antibody-based immunotherapies and antibody-conjugated nanoparticle-based targeted therapies, targeting specific tumor-associated antigens (TAA), show promise as potential innovative treatments.

Summary: This study found that a blood-brain barrier-permeable compound called 2,3-benzodiazepin-4-one (1g) can interfere with the activity and expression of brain glycogen phosphorylase (PYGB) in glioblastoma multiforme (GBM) cells, and inhibit cell growth and cycle. These findings suggest that PYGB may be a potential therapeutic target for GBM, and 1g has the ability to regulate glucose uptake and metabolism.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Summary: The genomic landscape of glioblastoma and its components, such as the stem cell compartment and peritumoral brain zone, is still incomplete. Research has shown the key role of stem cells in tumor maintenance and drug resistance, as well as the importance of the marginal area where tumor recurrences often occur. By analyzing the genomic profiles of different components, new specific biomarkers have been identified for potential therapies.

Summary: This study compared the efficacy of anti-PD-1 and anti-PD-L1-based therapy in R/M HNSCC patients through a systematic review and network meta-analysis. It found no significant differences in overall survival between different subgroups, except for metastatic patients where anti-PD-1-based therapy was associated with significantly less risk of death.
