The Role of Macrophages in the Pathogenesis of Atherosclerosis

Summary: This review summarizes the involvement of various cell populations, including immune cells and endothelial cells, in the pathogenesis of atherosclerosis. Among them, macrophages are of particular importance due to their diverse effects on atherogenesis and inflammation in atherosclerosis. The significance of different phenotypes of macrophages in the progression or regression of atherosclerosis is discussed, including their ability to become foam cells, the consequences of this event, and their ability to create a pro-inflammatory or anti-inflammatory environment through cytokine production at the site of atherosclerotic lesions. In addition, therapeutic strategies aimed at modulating macrophage activity are considered, providing useful ideas for future drug developments.

The early development of atherosclerotic lesions involves the accumulation of lipoprotein-associated lipids and their interaction with macrophages. Macrophages in early plaques engage with low-density lipoproteins (LDL), ingesting large amounts of cholesterol, which is then stored in lipid droplets. Hydrolysis of cholesteryl esters occurs extracellularly, leading to the formation of unesterified (free) cholesterol. High concentrations of cholesterol can also result in the formation of cholesterol hydrate crystals, observed in atherosclerotic blood vessels. Understanding this process may lead to novel therapies for atherosclerosis prevention and treatment.

The phagocytic role of macrophage following myocardial infarction

Summary: Myocardial infarction (MI) is a cardiovascular disease that has a high incidence and mortality rate. The disease results in a large number of apoptotic and necrotic cells that need to be efficiently and quickly engulfed by macrophages to prevent further necrosis. Phagocytic macrophages can reduce or resolve inflammation and protect the infarcted heart. The phagocytosis of macrophages is influenced by various factors such as proteins, receptors, lncRNA, and cytokines. Understanding the mechanisms of phagocytosis can help regulate macrophage phagocytic ability for cardioprotection after MI. This review provides an overview of the phagocytic effect of macrophages and summarizes the latest reported signals that regulate phagocytosis after MI, offering new insights into phagocytosis-dependent cardiac protection after MI.

The results showed that DAPA could attenuate inflammation by inhibiting the expression of TLR-4 and activation of NF-kappa B, as well as shifting macrophage polarization from an inflammatory state to an anti-inflammatory state.
